Immunofluorescence Analysis of Renal Fibrosis

The renal tissue sections were deparaffinized and rehydrated in an alcohol gradient and xylene and then boiled in citrate buffer for 20 min for antigen retrieval. After blocking with 5% goat serum for 30 min, the sections were incubated with primary antibodies: anti-fibronectin (FN, ab2413, Abcam, Cambridge, UK) and anti-collagen-IV (col-IV, ab6586, Abcam) overnight at 4°C. The cells were fixed in 4% paraformaldehyde for 15 min at room temperature. Primary antibody to PAL (rabbit anti-PAL) was used at 1:500 dilution. The secondary antibody, Alexa Fluor 594 anti-rabbit (Proteintech), was used at a concentration of 1:200.
The liver samples were removed from mice immediately upon sacrifice and fixed in 0.1 M phosphate buffer (pH 7.4) containing 4% paraformaldehyde overnight at 4°C, and then embedded in paraffin and sectioned.
