Visualizing Soluble SFRP2 Protein in D2.0R Cells

To visualize soluble SFRP2 protein, confluent D2.0R cells were cultivated in DMEM without serum. After 5 days, conditioned medium was pooled from three 15cm dishes/condition, spun 20’ at maximum speed to remove debris and then concentrated by spinning the samples for 30’ at 4°C at 3000rcf (Amicon Ultra-15 Centrifugal Filter Devices 30,000 MWCO, Millipore). As loading control, remaining cells were harvested and processed as in40 (link). Western blotting was performed as in40 (link). Antibodies: SFRP2 (1:1000, Abcam, ab137560), GAPDH (1:25000, Millipore, MAB374). Antibody for SFRP2 has been validated with recombinant mouse SFRP2 (R&D, 1169-FR).
