Establishment of Human Hepatocellular Carcinoma Cell Lines

PLC/PRF/5 and HepG2 human HCC cell lines were purchased from the National Institute of Biomedical Innovation JCRB Cell Bank (Osaka, Japan) and cultured with Dulbecco’s Modified Eagle’s Medium (DMEM). All cell culture supplements were purchased from Sigma-Aldrich (Milan, Italy).

Recombinant Insulin-like Growth Factor 1 (IGF1) was purchased from Calbiochem (San Diego, CA, USA) and recombinant Vascular Endothelial Growth Factor from ORF Genetics (Kopavogur, Iceland). The two cell lines were grown in monolayer culture with 10% fetal bovine serum (FBS), 100 U/mL penicillin and 100 μg/mL streptomycin addition and incubated at 37 °C in a humidified atmosphere containing 5% CO2 in air.

Evaluation of IGF1R Inhibitors and Doxorubicin

The IGF1R inhibitors OSI-906, NVP-ADW742 and GSK1838705A were purchased from Selleck Chemicals LCC (see Additional file 1: Table S1 for properties) and dissolved in DMSO in a concentration of 10 mM. The IGF1R inhibitors were tested in concentrations up to 1 μM as it was demonstrated previously that higher concentrations lead to an aspecific toxic response [24 (link)]. Recombinant human IGF1 (PeproTech) was used in a concentration of 50 ng/ml [25 (link)]. Doxorubicin was obtained from the in-house hospital pharmacy in a 0.9 % NaCl solution, and used in a concentration range of 1–100 nM.

GSK1838705A Inhibition of U87MG Glioma in Mice

GSK1838705A was purchased from Selleck Chemicals LLC (Houston, TX, USA). The human U87MG glioma cell line was purchased from American Type Culture Collection (Rockville, MD, USA). The U87MG cells were maintained in Eagle's minimal essential medium (EMEM), supplemented with 10% heat-inactivated fetal bovine serum (FBS) and 100 U/ml penicillin-streptomycin in a humidified incubator at 37°C with 5% CO2. Athymic nude mice were purchased from Slack Company (Shanghai, China) and were handled in compliance with the Experimental Animal Care and Use Committee guidelines of Xinxiang Medical University (Xinxiang, Henan, China). A total of 18 female mice (age, 7–8 weeks; weight, 24–28 g) were housed in a specific-pathogen-free facility maintained between 20 and 25°C with a 12 h light/12 h dark cycle, and were allowed ad libitum access to food and water. The study was approved by the ethics committee of Xinxiang Medical University (Xinxiang, China).
