Analyzing CRC Tumor RNF215 Expression

Statistical analyses were conducted with R software (version, 3.6.3) and Graph Prism (version, 9.0; GraphPad Software, Inc.; Dotmatics). The Wilcoxon rank sum test was used to compare the expression of RNF215 in CRC tumors with that in normal tissues. The Wilcoxon rank sum/Kruskal-Wallis test and logistic regression were performed to demonstrate the association between CRC clinicopathological characteristics and RNF215 expression. The CRC clinicopathological features correlated with survival were analyzed using the Kaplan-Meier method and Cox regression (23 (link)). Multivariate Cox analysis was conducted to assess the effect of RNF215 expression and the other clinicopathological features on survival. The variables with P<0.1 in univariate Cox regression analysis were further assessed in multivariate Cox regression analysis. Two-sided P-values <0.05 were considered to indicate a statistically significant difference.

Statistical Analysis of Survival Data

Microbial Diversity Analysis Pipeline

Sequence analysis was performed using Uparse software (Uparse v7.0.1001). Sequences with ≥97% similarity were assigned to the same OTUs. To study the phylogenetic relationships of different OTUs and the differences between the dominant species in different groups, multiple sequence alignment was conducted using MUSCLE software (version 3.8.31). Alpha diversity was applied to analyze the complexity of the species diversity for a sample through three indices, including Chao1, Shannon, and good coverage. All these indices in our samples were calculated with QIIME (version 1.7.0) and visualized with R software (version 2.15.3). The beta diversity based on weighted UniFrac was calculated using QIIME software (version 1.9.1). Random forest analysis evaluates the importance of each predictor by determining how much the mean square error (MSE) increases. The variables were selected when the predictor variables were randomly replaced and the other variables remained unchanged. These analyses were performed using the “RandomForest” package in R. The “Hmisc” package was used to calculate the correlation, the “igraph” package was used to analyses the co-occurrence network (Csardi and Nepusz, 2006 ), and Gephi 0.9.2 software was used to draw the network map. Cytokine Levels in VitD-Treated COVID-19

Plasma levels of IL-17, IL-6, IL-1β, and TNFα in VitD-treated and untreated patients was adjusted for patient's age, male sex, body mass index (BMI), and comorbidity of diabetes mellitus (DM) using a linear regression model. The length of ICU stay between these treatment groups were assessed using a generalized linear mixed model, which included fixed effect of treatment and random effect of age, male sex, BMI, and DM. Statistical analysis was performed using IBM SPSS version 26.0, R software version 3.6.1, and GraphPad Prism version 8.0. Furthermore, comparison between two groups was done using unpaired t-test or Mann-Whitney U test, depending on the skewness of the data. All tests were two-tailed and a P value of <0.05 was considered statistically significant.
