Synthesis and Characterization of Lyso-7 Compound

Lyso-7, [(5Z)-5-[5-bromo-1H-indol-3-yl)methylene]-3-(4-chlorobenzyl)-thiazolidine-2,4-dione] (CAS Number 1505484-42-3) (CAS Registry Number 1505484-42-3), Mw 447.73 g/mol, was synthesized, purified and characterized by Prof Ivan da Rocha Pitta as reported [11 (link)]. PLA polymer, a poly-rac-lactide Mn 75,000–120,000 g/mol, poloxamer 188, polysorbate 80, polyethylene glycol 300 (PEG 300), glucose, ethanol and methanol (HPLC grade) were purchased from Sigma-Aldrich (Cotia, Brazil). Symplicity® System (Millipore, Bedford, MA, USA) produced Milli-Q water to prepare all solutions throughout the experiments. Soy lecithin with 75% of phosphatidylcholine (Epikuron® 170) was a generous gift from Lucas Meyer (Champlan, France). Sasol Olefins & Surfactants GmbH (Hamburg, Germany) provided Miglyol 810 N. Tedia (Rio de Janeiro, Brazil) provided ethyl acetate, acetone, N,N-dimethylacetamide (DMA), acetone and dimethylsulfoxide (DMSO) (analytical grade).

Nanocarriers for Targeted Delivery

All reagents for culture cells were obtained from Life Technologies SARL (Cergy Pontoise Cedex, France). Epikuron® 170 (~70 % soy phosphatidylcholine) was a gift from Lucas Meyer (Le Blanc Mesnil, France). Miglyol® 810 N oil (capryc/caprylic triglyceride) was purchased from Hülls (Frankfurt, Germany). The poly-ε-caprolactone (PεCL, average 42,500 Da), Poloxamer®188, DHA, α-tocopherol (vitamin E) and hydrogen peroxide (H2O2) were provided by Sigma-Aldrich (St. Louis, MO) and the acetone was purchased from Mallinckrodt chemicals (USA). Symplicity® System (Millipore, Bedford, USA) was used to produce Milli-Q water (18.2 MΩ), which was used to prepare nanocapsules and all solutions for this study. The synthesis of 4(RS)-4F4t-neuroprostane was performed according to our published procedure [47 ].

Benznidazole (BZ) (N-benzyl-(2-nitro-1H-imidazol-1-yl), Mw 260.25 g/mol, purity 99.85% (2) (Fig. 1), Poloxamer ® 188 and polymer poly-rac-lactide (PLA), Mn 75,000-120,000 g/mol, Tween ® 80, Polyethylene glycol 300 and methanol (HPLC grade) were purchased from Sigma-Aldrich (Brazil). Symplicity ® System (Millipore, Bedford, USA) was used to produce Milli-Q water, which was used to prepare all solutions throughout the experiments. Soy lecithin (Epikuron ® 170) was provided by Lucas Meyer (France) as a kind gift. Mygliol 810 N was provided by Hulls (Germany). Ethyl acetate, N,N-dimethylacetamide (DMA) and dimethylsulfoxide (DMSO) (analytical grade) were provided by Tedia (Rio de Janeiro, Brazil).
