BV2 Microglia Cell Culture Protocol

BV2 microglia cells were obtained from GuangZhou Jennio Bio- tech. HT22 Cell Culture and Compound Treatments

Cobalt Chloride and Propofol Treatment of PC3 Cells

A 1-mM stock solution of cobalt chloride (CoCl2) was prepared by dissolving CoCl2 powder (Sigma-Aldrich, Dorset, UK) in dH2O, this stock solution was then further diluted to a concentration of 100 μM with serum-free propofol (0–4 μg ml−1 in intralipid)-supplemented PC3 medium. Before treatment, PC3 cells were cultured at a density of 1 × 106 per ml on 30-mm2 Petri dishes, as described above. Complete medium was then replaced with CoCl2 and propofol-supplemented medium for 6 h. Cells were harvested immediately after exposure for western blotting.
