HDAC Activity Determination Assay

HDAC1 (BML-AK511), HDAC6 (BML-AK516) and HDAC8 (BML-AK518) activity was determined using a commercial assay kit from Enzo Life Sciences (HDAC fluorometric assay/drug discovery kit, Farmingdale, NY, USA). The assay was carried out according to the manufacturer’s instructions. Fluorescence intensity was measured at an excitation wavelength of 360 nm and an emission wavelength of 460 nm using a fluorometer (LS 55, Perkin Elmer, USA). Trichostatin A (TSA) was also evaluated as a positive control. IC50 values were determined using GraphPad Prism 5 software.
