Antioxidant Evaluation of DPPH Assay

ammonium thio-cyanate, ascorbic acid (vitamin C), aluminum chloride
(AlCl3), ferrous chloride, potassium ferri-cyanate, sodium
nitrite, ferric chloride, butylatedhydroxytoluene (BHT) (99.0 %),
butylatedhydroxyanisole (BHA), dimethyl sulfoxide,
3-(4,5-dimethylthiazol-2,5-diphenyl tetrazolium bromide) (MTT), penicillin/
streptomycin solution, and fluconazole were supplied by Sigma Chemical
Corporation, USA. Sterile resazurin tablets for the purpose of sterilization

ammonium thiocyanate, ferrous chloride, methanol, and sodium carbonate
(anhydrous) used in the research were acquired from Merck, Germany, unless
stated otherwise. Analytical grade reagents and chemical were used for this
research work.

Fish Oil Characterization and Analysis

Fish oil (35% PUFA) containing 17% EPA, 11% DHA and 0.12% free fatty acids. The peroxide value (PV) of oil was 1.3 equivalent of peroxides/kg of oil, and anisidine value was 13.9 according to the specifications of the manufacturer) was purchased from Alhavi Company (Iran). The fish oil was stored in a freezer at 75°C until use. Tween 20, tween 60, tween 80, lecithin, sodium dodecyl sulfate (SDS), poloxamer 407, Precirol®ATO5 (glyceryl palmitostearate), stearin, palmitic acid, 1,1,3,3-tetraethoxypropane and α-tocopherol were provided from Sigma Aldrich (Steinheim, Germany). Chloroform, methanol, ammonium thiocyanate, Iron (II) and cumene hydroperoxide were purchased from Merck (Germany). All materials were used without further purification. Also, ultra-pure water was used throughout the study.

Detection of γH2AX in Skin Samples

Detection of γH2AX in paraffin-embedded sections was facilitated by antigen retrieval in citrate buffer pH = 6. Cytospins with epidermal cells were fixed in 4% formaldehyde for 15 min at room temperature. Incubation with anti-γH2AX or isotype control was performed overnight at 4°C after blocking with 10% goat serum for 1 h at room temperature. Antibody binding was visualized using anti-rabbit Alexa Fluor 594 Ab. Antibodies were diluted in 2% goat serum or BSA containing 0.3% or 0.5% Triton X-100. Cytospins were counterstained with anti-MHC class II-FITC mAb. DAPI was used to counterstain cell nuclei. γH2AX+ cells in paraffin-embedded sections were counted with a calibrated grid (7–10 fields per sample). Epidermal sheets were prepared by separation with ammonium thiocyanate (Merck, Westchester, PA, USA), fixed with acetone and stained with anti-MHC class II-FITC mAb (30 (link)). Staining was visualized with a 10×, 40× or 100× objective using an Olympus BX60 epifluorescence microscope (Hamburg, Germany).

Extraction and Characterization of Nettle-Derived Biomaterials

NMR, Chromatography, and Spectroscopy Methods

Cobalt Extraction and Detection Protocol

The salt of CoCl2.6H2O (Merck company, 98.0%) was acquired for the preparation of cobalt solution. Bis (2,4,4-trimethylpentyl monothiophosphinic acid) (Cyanex302) (Aldrich company, 90.0%), bis(2,4,4-trimethylpentyl)dithiophosphinic acid (Cyanex301) (Aldrich company, 90.0%), bis-(2,4,4-trimethylpentyl) phosphinic acid (Cyanex272) (Aldrich company, 90.0%), and Di-(2-ethylhexyl)phosphoric acid (D2EHPA) (Merck company, 95.0%), as the extractants and kerosene (99.9%) as the diluted phase from Merck company, were used in the experiments. Ethylene glycol from Sigma Aldrich, 99.5% was used as the non-aqueous system. Another diluted phase such as, chloroform (99.8%), 1-decanol (99.0%), and toluene (99.9%) was provided from Merck. HCl (37.0%, Merck) and NaOH (99.9%, Merck) solutions were used to reach the appropriate pH of solutions in the experiments. As reverse extraction agents, acids like sulfuric acid (96.0%, Merck), nitric acid (65.0%, Merck), and hydrochloric acid were utilized. Ascorbic acid (99.0%, Merck), ammonium thiocyanate (99.0%, Merck), acetone (99.0%, Merck), and hydrochloric acid were used as cobalt detectors using a UV spectrophotometer.
