Analytical Standards of 20 Amino Acids by HPLC

The analytical standards of 20 amino acids, including aspartic acid (Asp), glutamic acid (Glu), γ-am aminobutyric acid (GAMA), serine (Ser), glutamine (Gln), histidine (His), asparagine (Asn), glycine (Gly), threonine (Thr), arginine (Arg), alanine (Ala), tyrosine (Tyr), valine (Val), methionine (Met), tryptophan (Trp), phenylalanine (Phe), isoleucine (Ile), leucine (Leu), lysine (Lys), and proline (Pro), were obtained from Sigma-Aldrich Co. (St. Louis, Mo, USA) with all purity ≥ 99%. Phenyl isothiocyanate (PITC) (purity ≥ 99%) and triethylamine (purity ≥ 99%) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Ammonium acetate acetonitrile (HPLC grade) and hexane (HPLC grade) were obtained from Dikma (Beijing, China). Ultra-pure water was prepared using a Milli-Q plus system (Millipore, Boston, Massachusetts, The United States of America).
The individual stock solutions were prepared in 0.1 mol/L HCl solution at the concentration of 10,000 μg/mL. The standard mixture was prepared from the individual stock standard solutions at the concentration of 100 μg/mL. The working solutions of the amino acid were prepared by serial dilutions. The internal standard norleucine was prepared with a concentration of 100 μg/mL. All the solutions were stored at 4 °C in the refrigerator.

Quantitative Analysis of Amino Acids in Diverse Soy Sauces

Seafood soy sauce (Haitian), jinbiao soy sauce (Haitian), steamed fish soy sauce (Lee Kum Kee), spicy fresh dew (Knorr), fresh shellfish sauce (Totole), Maggi fresh (Nestle), celery, carrot, onion, green onion, red pepper, ginger, garlic, coriander, dried shrimps, dried shitake mushrooms, and rock sugar were purchased from local supermarkets in Wuxi, China. Food-grade ascorbic acid and potassium sorbate were purchased from Henan Wanbang Industrial Co., Ltd. (Zhengzhou, China). The juicer was purchased from Midea Group Co., Ltd. (Foshan, China).
Methanol (chromatographically pure) and acetonitrile (chromatographically pure) were purchased from Tedia Co., Ltd. (Fairfield, Ohio, USA). Tetrahydrofuran, triethylamine, hydrochloric acid, crystalline sodium acetate, and trichloroacetic acid were of analytical grade, and water was Millipore ultrapure water. Seventeen amino acid standards, including alanine (Ala), arginine (Arg), glycine (Gly), glutamic acid (Glu), aspartic acid (Asp), methionine (Met), serine (Ser), histidine (His), leucine (Leu), phenylalanine (Phe), isoleucine (Iso), cysteine (Cys), tyrosine (Tyr), lysine (Lys), threonine (Thr), valine (Val), and proline (Pro), were purchased from Sigma-Aldrich (Shanghai) Trading Co., Ltd. (Shanghai, China).

Plasma Amino Acid Analysis by HPLC

500 uL of plasma from each animal was precipitated with ammonium sulfate up to 60%, to remove the globulin, remaining over-night in the refrigerator. After centrifugation (3400 rpm, 5 min, eppendorf), precipitates were resuspended in phosphate buffer pH = 7 and dialyzed (Tubbing MW 10,000 daltons) for the removal of the salt used. Samples were filtered in millipore 0.25 um, and 20 uL was injected into HPLC. YL-9300 UV-coupled HPLC instrument 254 nm, C18 Luna 25 cm × 4.5 mm reverse phase column, temperature 27–30 °C. Separation was done in the following mobile phase: buffer sodium acetate 10 mM in MilliQ water (A) and acetonitrile (B): 0–1 min (20% B); 1.01–1.5 min (5% B); 1.51–8 min (4% B). The flow rate was kept constant at 1 mL/min and peaks were detected at 254 nm. All chemicals reagents used in the analysis, such as acetonitrile and acetate buffer we purchased from Sigma and Merck, (Rio de Janeiro Brazil), with high purity levels. The standards used were Kynerunin-Kyn (20 uM), Tryptophan-Trp (20 uM), Cysteine-Cys (10 mM), Arginine-Arg (10 mM), Metionine-Met (10 mM), Proline-Pro (10 mM), Valine-Val (10 mM), Alanine-Ala (10 mM), Phenilalanine-Phe (10 mM), Tyrosine-Tyr (10 mM) from Sigma and Merck. All were evaluated and separated by HPLC based on the peak at said retention time (Rt) and height peak in millivolt (mV) or concentration in uM or ug.
