Saponin was from Merck (Darmstadt, Germany), Zymozan A, albumin, DMSO and glycerol from Sigma (St. Louis, MO, USA), sterile endotoxin-free Hank’s balanced salt solution (HBSS) from Thermo Scientific (Waltham, MA, USA) and Ficoll from Phadia (Uppsala, Sweden). All other reagents were of research grade. HBSS contained 135 mM NaCl, 4 mM KCl, 1 mM MgCl2, 2 mM CaCl2, 10 mM glucose, 1 mM Na2HPO4, 20 mM HEPES and pH=7.3. Anti-inflammatory Compound Evaluation

Carrageen was purchased from FCM Corporation (USA), Zymozan A and Evans blue from Sigma-Aldrich (Poland), Caffeine (used as standard in obese model) was purchased from Alfa-Aesar (Poland). KD-64—(1,3-dimethyl-9-((1r,4r)-4-methylcyclohexyl-6,7,8,9-tetrahydropyrimido[2,1-f]purine-2,3(1H,3H)-dione) (Fig 1B) [43 (link)] was synthesized in the Department of Technology and Biotechnology of Drugs, Faculty of Pharmacy, Jagiellonian University Medical College, Cracow, Poland. NMR and LC-MS techniques assessed identity and purity of final product. In all experiments Ketoprofen, Caffeine or KD-64 were administered as suspensions in 1% Tween 80 in sterile water for injection.
