Tissue Preparation and Immunostaining Protocol

To generate tissues for immunostaining, mice were transcardially perfused with DPBS to remove blood, then perfused with 4% paraformaldehyde in PBS, the tissues were dissected and cryopreserved in 30% sucrose, frozen in 2:1 30%sucrose:OCT and then 10 micron sections were generated. For immunostaining, tissue sections were blocked/permeabilized with 0.1–5% Triton X-100 and 10–50% goat serum in PBS. Tissue sections were incubated in primary antibodies overnight at 4°C and secondary antibodies for 1.5 hours at room temperature with 3% goat serum, 0–0.1% Triton X-100 in PBS.
