Western Blot Analysis of Extracellular Matrix Proteins

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total cell lysates were separated by 8% or 10% SDS-PAGE and then electroblotted and transferred onto PVDF membranes.31 (link) After incubation with primary antibodies specific for COL1A1 (1:1000; sc-293182, Santa Cruz, TX, USA), CD44 (1:2000; #37259, CST, Boston, USA), E-cadherin (1:2000; #14472, CST, Boston, USA), vimentin (1:2000; #5741, CST, Boston, USA), TGFBI(1:1000; 10188–1-AP, proteintech, IL, USA), MMP9 (1:2000; #15561, CST, Boston, USA) and β-actin (1:4000,sc-81178, Santa Cruz, TX, USA) at 4 °C overnight, the blots were incubated with the corresponding secondary antibodies at room temperature, and the proteins were detected with an ECL kit.

Western Blot Analysis of Key Proteins

Check if the same lab product or an alternative is used in the 5 most similar protocols
The whole cell lysates were harvested and subjected to SDS-PAGE (10% separation gel) and transferred to a polyvinylidene difluoride (PVDF) membrane. Primary antibodies against the following proteins were used: HMGB1 (10829-1-AP; Proteintech), TGFBI (10188-1-AP; Proteintech), GAPDH (10494-1-AP; Proteintech), and actin (60008-1-Ig; Proteintech). Horseradish peroxidase–conjugated secondary antibodies were used (ZSGB-BIO). Signals were detected using an ECL (enhanced chemiluminescence) kit (Millipore).

Immunohistochemical Analysis of HGSOC Biomarkers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immunohistochemistry (IHC) analysis was conducted on samples collected from 41 HGSOC patients. It was done according to standard protocols using antibodies against IGFBP4 (ProteinTech), TGFBI (ProteinTech), and serpin family A member 10 (SERPINA10) (ProteinTech); and cytoplasmic staining was considered as positive. Staining was assessed by three independent pathologists (WG, XH, and YW). The immunoreactive score (IRS) gave a range of 0–12 as a product of multiplication between positive cell proportion score (0, no positive cells; 1, <10% of positive cells; 2, 10–50% positive cells; 3, 51%–80% positive cells; and 4, >80% positive cells) and staining intensity score (0, no color; 1, mild staining; 2, moderate staining; 3, intense staining) (13 (link)).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
