Cell culture supernatant (100 µL) was added to a microwell plate coated with an anti-human polymorphonuclear (PMN) elastase polyclonal antibody and incubated at room temperature for 1 hour with a Horseradish peroxidase-conjugated anti-α 1-proteinase inhibitor antibody. The immune complex was detected by adding tetramethyl-benzidine substrate solution, and the absorbance was determined at 450 nm on a microplate reader (BioTek Synergy H1 Hybrid).
