DOTA-WVP Peptide Synthesis and Characterization

WVP peptide was manufactured using the solid-phase peptide synthesis method by ChinaPeptides Co., Ltd. (Shanghai, China). During the synthesis process, the C-terminus of WVP was modified with DOTA to obtain DOTA-WVP. Sodium acetate (NaOAc), hydrogen chloride (HCl), 3-aminopropionitrile fumarate salt (BAPN), Sirius red, and picric acid were supplied by Sigma-Aldrich (St. Louis, MO, USA). Neutrophil Quantification in Frozen Tissue

Goat-anti-mouse S100A9 (R&D, 100ng/ml) and rabbit- anti-mouse colIV (Abcam, 13.3μg/ml) immunofluorescence staining was performed on frozen tissue, followed with fluorescently conjugated secondary antibodies (chicken anti-goat AlexaFluor488 and donkey anti-rabbit AlexaFluor647, Invitrogen, 10μg/ml). Nuclei were visualized using DAPI. Images were acquired using a Leica SP5 DM confocal microscope. Neutrophils were manually counted in each 400× field.
