Effect of pH on Staphylococcus aureus Growth

Normal TSB medium was adjusted to have a pH of 7.4. TSB culture mediums with different pH values (8, 8.5, 9, and 9.5) were prepared by adding a 1 M NaOH solution dropwise to normal TSB while monitoring the mixture with a pH meter (Mettler Toledo320, Zurich, Switzerland). The Staphylococcus aureus (ATCC25923) bacteria were inoculated in TSB medium with different pH values (7.4, 8, 8.5, 9, and 9.5) in 48-well plates at a density of 1 × 106 CFUs/mL. Then, the culture plates were incubated at 37 °C for 360 and 480 minutes. At each time point, the incubated bacterial suspensions were blended, plated onto TSA, incubated for 24 hours, and counted.
