Western Blot Analysis of Chromatin Regulators

Whole-cell extracts were obtained from cells washed with PBS, collected with Laemmli buffer (4% [vol/vol] SDS, 20% [vol/vol] glycerol, and 120 mM Tris-HCl, pH 6.8), and then sonicated with a Diagenode Bioruptor for 10 min followed by 5 min of boiling at 95°C before loading. Samples were resolved by SDS-PAGE and analyzed by standard WB protocols. Signals of the Western blots were detected by standard chemiluminescence (GE Healthcare) and analyzed using a ChemiDoc XRS+ system (Bio-Rad Laboratories).
