Protein Expression Analysis in Cell Lysates

Cell lysates were harvested as previously described [16 (link)]. Cytoplasmic protein was extracted using the P0013G extraction kit (Beyotime, Shanghai, China), according to manufacturer's protocol. Protein extracts were isolated through SDS-PAGE gel electrophoresis and were then transferred to a PVDF membrane. Membranes were incubated with primary antibodies against FYN, CDH1, VIM, SNAI1, SNAI2, phosphorylated-STAT3, and STAT3 (Cell Signaling Technology, Danvers, MA; USA, 1:1000). Membranes were also incubated with a primary antibody against GAPDH (Sigma-Aldrich, St Louis, MO, USA, 1:2000). Images were obtained using the Image Lab Software (Bio-Rad, Hercules, CA, USA).
