Colorectal Cancer Cells miR-199a-5p Modulation

All colorectal cancer cells (DLD-1, HT29, ANU-C2B, SNU-C4, SNU-C5, Colo320 DM)
were purchased from the Korean Cell Line Bank and maintained in RPMI-1640 Medium
(Invitrogen) containing 10% fetal bovine serum with 5% CO2 in a
37℃ incubator. miR199a5p mimics and negative mimics were purchased from
Dharmacon. Cells were transfected with a miR-199a-5p mimic using the DharmaFECT
1 transfection reagent (Dharmacon) according to the manufacturer’s
protocol. The negative mimic was used for control purposes. After 72 h of
incubation, cells were harvested and used for extraction of total RNA or
protein.

Establishment and Characterization of Human CRC Cell Lines

The human CRC cell lines Caco-2, COLO-320DM, DLD-1, HCT-8, HCT-15, HT-29, LoVo,
SW480, SNU-C2B, SNU-C5, and SNU-175 were purchased from the Korean Cell Line Bank
(Seoul, Korea). The DiFi cell line was kindly provided by Dr. J. O. Park (Samsung
Medical Center, Seoul, Korea). The KRAS mutation status of all cell
lines are summarized in Table 1[12 -15 (link)]. All cell lines were
cultured in RPMI-1640 medium (Gibco, Grand Island, NY), supplemented with 10% fetal
bovine serum (FBS; Gibco) and 1% penicillin/streptomycin solution (WelGENE Inc.,
Daegu, Korea), and were maintained at 37°C in 5% atmospheric
CO2.

Cell Lines for Colorectal Cancer Research

Human CRC HCT 116, HCT-15, COLO 320DM, HT-29, DLD-1, Caco2, SW480, SW480E, SW620, and RKO cell lines were purchased from the Korean Cell Line Bank (KCLB, Seoul, Korea). Human embryonic kidney 293T cells were obtained from American Type Culture Collection (ATCC, Manassas, VA, USA). Cell lines were cultured in RPMI 1640 (Welgene, Daegu, Korea) or DMEM (Welgene) medium containing 10% heat-inactivated fetal bovine serum (Gibco, Thermo Fisher Scientific, Waltham, MA, USA) at 37°C in an incubator with a humidified atmosphere of 5% CO2.
