Ethical Animal Experiments in Dental Research

The Animal Care Committee of Fukuoka Dental College approved all animal procedures used
in this study. The protocol for these experiments was reviewed and approved by the
committee of Ethics of Animal Experiments of Fukuoka Dental College (#18008). All mouse
experiments were performed in accordance with the guidelines of the committee of Ethics of
Animal Experiments of Fukuoka Dental College. C57BL/6N (B6) mice (CLEA Japan, Shizuoka,
Japan) and IL-17A-GFP reporter mice (The Jackson Laboratory, Bar Harbor, ME, USA) were
kept under specific pathogen-free conditions in the animal facility of Fukuoka Dental
College. All animals were kept in a controlled environment with a 12 h light/dark cycle
(lights on at 7:00 am). Mice were group housed (2–5 per cage) and given access to food and
water ad libitum.

Genetic Manipulation of Lgals8 in Mice

Mice were mantained under conditions of strict confinement, which included automatic control of temperature (21°C) and photoperiod (12 h light / 12 h dark). Lgals8/Lac-Z knock-in (here called Gal-8 KO or Lgals8-/-) mice were generated from C57BL/6NTac mice engineered in Regeneron Pharmaceuticals Inc., New York, using Velocigene technology for replacing the entire coding region of the mouse Lgals8 gene (18,427 bp) with LacZ lox-Ub1-EM7-Neo-lox Cassette containing the LacZ gene that encodes β-galactosidase [40 (link)]. Details of the Lgals8 KO mice and PCR genotyping assay, including the predicted PCR products and the primers, are available at the Velocigene website (www.velocigene.com/komp/detail/14305). IL-17A-GFP reporter mice in the C57BL/6J background, which express EGFP under the control of the IL-17A promoter, were purchased from Jackson Laboratories (Bar Harbor, ME).
