the blood glucose level was determined by glucometer (Accu-Chek Performa, Roche Diagnostics). Urine samples were centrifuged at 4 °C and 2000 rpm for 10 min. The urinary albumin and creatinine concentrations in the supernatant were measured using Albumin Rat ELISA kit (Abcam, UK) and creatinine ELISA kit (Cusabio, China) according to the manufacturer’s protocols. The urinary albumin to creatinine ratio (UACR) was calculated based on the albuminuria and urine creatinine level (albumin mg/creatinine g) [16 (link)]. Serum progesterone and estradiol levels were measured by commercially available ELISA kits (Biocheck, Inc. Foster City CA, USA and Cusabio, wuhan, China, respectively), according to the manufacturer’s protocols.

Evaluating Nephroprotective Effects of ZnONPs

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 7 weeks of ZnONPs treatment, rat urine collection was performed using individual metabolic cages for 24 h. Urinary albumin concentration was measured using the Albumin Rat ELISA kit (cat no. ab235642, Abcam, UK) according to manufacturer instructions. Using commercially available kits, blood glucose and blood urea nitrogen levels (BUN) were measured (ARCOMEX, Jordan). The rat kidney weight (mg) to body weight (g) ratio was calculated to obtain the kidney weight index (KWI).

Renal Function Assessment Post-Ablation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Three and six weeks after renal ablation, the rats were placed in metabolic cages (Nalgene, Rochester, NY, USA) to collect 24-h urine. Urine samples were centrifuged at 5000× g for 15 min to remove debris, and the supernatant was analyzed. The variables measured were creatinine and blood urea nitrogen (BUN) (IL 300 plus, Clinical Chemistry Analyzer, Instrumentation Laboratory, Bedford, MA, USA), microalbuminuria (Albumin Rat ELISA Kit, Abcam Cambridge, MA, USA), and urinary N-acetyl-β-d-glucosaminidase (NAG) activity (spectrophotometric assay).

At 4 weeks of age, age-matched male SS-WT (n=9) and SS-Arhgef11−/− (n=10) were weaned onto a low-salt diet (0.3% NaCl). Twenty-four-hour urine collections were done at 6 weeks of age on low salt and then each group was placed on high salt (2.0% NaCl) for the remainder of the study. Albuminuria was measured using a rat albumin ELISA kit (Abcam, ab108789). At week 7, a subset of animals from each group (n=8) were implanted with telemetry transmitters (model HD-S10, Data Sciences International) as previously described.13 (link),16 (link) After 1 week of recovery, blood pressure was measured for 24 hours one day each week from weeks 8 to 15, followed by 24-hour urine collections were made for the determination of proteinuria and albuminuria. Euthanasia was performed after week 15 blood pressure, and urine collections were complete. Organ weights, serum, and kidney samples for histological examination were collected.

Metabolic Biomarkers in Empagliflozin Treatment

Check if the same lab product or an alternative is used in the 5 most similar protocols
EDTA was added to blood samples followed by centrifugation (4,500 rpm) for 20 min at 4 ℃, then plasma was collected and stored at -20 ℃ until analysis. Urine samples were centrifuged (12,000 rpm) for 10 min at 4 ℃. The supernatant was frozen in liquid nitrogen until analysis. Levels of plasma creatinine, urea, glucose, and insulin as well as urinary creatinine, and total protein were detected using an automatic biochemistry analyzing system (Roche Cobas 6800, Roche Ltd, Switzerland). Levels of plasma BNP45 and urinary albumin were determined quantitatively using Rat BNP 45 ELISA Kit (Abcam, Cat#ab108816) and Rat Albumin ELISA Kit (Abcam, Cat#ab235642). The glomerular filtration rate-to-body weight ratio (GFR), albumin-to-creatinine ratio (ACR) were calculated. At a dose of 1 mg/day of empagliflozin urinary sodium and potassium excretion are not affected (data not shown).

Albumin and urea synthesis were quantified in spheroid media supernatant samples using Rat Albumin ELISA Kit (Abcam) and Urea Assay kit (Abcam) following the manufacturer's protocol. Supernatant samples were collected 4 days after media changes over the 21-day culture period. Data was normalised to account for cell number variation.

Evaluating Kidney Function Metrics in Rats

Check if the same lab product or an alternative is used in the 5 most similar protocols
All rats were housed in metabolic cages for 24 h (Pestel et al.) for blood and urine collection. After urine collection, blood samples were obtained from the lateral tail veins. Serum and urine creatinine levels were evaluated using a creatinine assay kit (ab65340; Abcam Plc, Cambridge, U.K.). The estimated glomerular filtration rate (eGFR) was obtained from creatinine clearance measurement in ml/min as follows: eGFR = (Ucr × V)/Pcr, Ucr = creatinine concentration in urine (mg/dL), V = urine flow rate (mL/min), and Pcr = creatinine concentration in plasma or serum (mg/dL)59 .
The albuminuria levels were evaluated using a rat albumin ELISA Kit (ab108789; Abcam Plc, Cambridge, U.K.). The albuminuria results are expressed in ng/mL. Thereafter, urine creatinine and urine albumin levels were calculated as the urinary albumin-to-creatinine ratio to indicate the level of kidney damage.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
