Cell Cycle Analysis of Synchronized Cultures

A total of 1×105 cells were treated with DMEM culture without FBS for 24 hours to synchronize cell cultures. The culture medium was then substituted with DMEM culture medium which contained 10% FBS. Cells were harvested after 48 hours and fixed in 75% ethanol at −20°C for 24 hours. Afterwards, cells were rinsed in PBS three times, rehydrated and resuspended in propidium iodide (PI)-RNase A solution (Invitrogen, USA) at 37°C for 30 minutes. Cell cycle was analyzed with a FACSCalibur flow cytometer (Biosciences, San Jose, CA, USA).
