Measuring ALT Activity in Mouse Serum

Check if the same lab product or an alternative is used in the 5 most similar protocols
ALT enzymatic activity was measured by an ALT activity assay (Sigma-Aldrich) in 10 µl of mouse serum according to the supplier’s instructions.

Comprehensive Metabolic Profiling of Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum insulin and glucagon levels were determined by the Mouse Ultrasensitive Insulin ELISA kit (ALPCO) and Mouse Glucagon ELISA Kit (Crystal Chem), respectively. In addition, the Serum Triglyceride Assay Kit (Abcam) and the ALT Activity Assay (Sigma) were used. Urine glucose levels were assessed using a Mouse Glucose Assay Kit (Crystal Chem).

Serum Collection and ALT Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum was collected from mice at euthanization, based on disease-burden. Whole blood was collected, allowed to sit at room temperature for 15 minutes, then centrifuged for 20 minutes at 4°C. The resulting serum was collected and stored at −80°C until use. ALT was measured using a commercially available ALT Activity Assay (Sigma-Aldrich), following the manufacturer's protocol.

Serum ALT Measurement in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Alanine aminotransferase (ALT) was measured in the serum of mice using the Vitros DT60 II chemistry system (Ortho-Clinical Diagnostics, Inc.) (Zheng et al., 2016 (link)) or ALT Activity Assay (Sigma) per manufacturer’s instructions. Method of ALT measurement was consistent within experiments.

Alanine Aminotransferase Activity Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Alanine Aminotransferase Activity (ALT) was assessed on the plasma of STD and MCD diet fed mice (at 4 and 8 weeks) with the ALT activity assay (Sigma-Aldrich), according to the manufacturer protocol. Results are express in International Unit (IU)/mL.

ALT Activity Assay for Liver Function

Check if the same lab product or an alternative is used in the 5 most similar protocols
Liver enzyme function was examined using the ALT Activity Assay (MAK052, Sigma-Aldrich). As a positive control, we used LPS to induce high levels of secreted ALT levels in the plasma as previously shown(Baveja et al., 2002 (link)).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
