PBMCs were surface stained with four panels of antibody cocktails (see Supplementary Table S2 ) for 15 minutes at 37°C. For panel 1, cells were then incubated with streptavidin APC for 10 minutes at 4°C. Cells were incubated with fixable viability dye eFluor 780 (Thermo Fisher Scientific) in PBS for 10 minutes at 4°C. For intracellular staining in panel 2, cells were fixed and permeabilized using the Foxp3/Transcription Factor Staining Buffer Set (Thermo Fisher Scientific) and incubated with an antibody cocktail containing anti-human Ki67 Alexa Fluor 488 (clone: Ki-67, Biolegend), CTLA-4 PE (clone: BNI3, BD Biosciences) and Foxp3 APC (clone: 236A/E7, Thermo Fisher Scientific) for 30 minutes at 4°C. Samples were acquired on a BD LSRFortessa (BD Biosciences) using the BD FACSDiva software v.8.0.2 (BD Biosciences).
