Human hematopoietic CD34+ cells from a 23 year old Caucasian male were procured from the National Hematopoietic Cell Processing Core, Fred Hutchinson Center Research Center (Seattle, WA, USA) [20 (link)]. Cells were cultured in serum-free medium consisting of Iscove’s Modified Dulbecco’s Medium (IMDM) supplemented with BIT 9500 (Stemcell Technologies Inc., Vancouver, BC, Canada), penicillin (100 U/ml; Invitrogen Corporation, Carlsbad, CA, USA), streptomycin (100 μg/ml; Invitrogen Corporation), recombinant human (rh) stem cell factor (SCF; 100 ng/ml; Stem cell Technologies Inc.), rh Fms like tyrosine kinase 3 (FLT3) ligand (100 ng/ml; Stemcell Technologies Inc.), and rh interleukin 3 (IL-3; 25 ng/ml; Stemcell Technologies Inc.). Culture flasks/dishes were incubated at 37°C with 5% CO2.
