Analytical Characterization of Phytochemicals

All the solvents, reagents, and standards used were of analytical grade. The following substances were used in the study: ethanol 96% (v/v) (AB “Vilniaus degtinė”, Vilnius, Lithuania), 4-(dimethylamino)cinnamaldehyde (99% purity), acetic acid (99.8% purity) (“Scharlau”, Sentmenat, Spain), procyanidin A2 (99% purity), procyanidin C1 (90% purity), quercetin (95% purity), quinic acid (98% purity), hyperoside (97% purity), isorhamnetin-3-O-rutinoside (95% purity), avicularin (90% purity), formic acid (97.5–98.5% purity), acetonitrile (99.9% purity), (+)-catechin (98% purity), rutin (95% purity), isoquercitrin (90% purity), chlorogenic acid (95% purity), and hydrochloric acid (37% purity) (Sigma-Aldrich, Steinheim, Germany). During the study, we used purified de-ionized water prepared with the “Milli–Q®” (“Millipore”, Bedford, MA, USA) water purification system.

Quantitative Analysis of Polyphenolic Compounds

Double-distilled water (ddH2O) was used in all experiments and samples for HPLC-DAD and LC-ESI-Q-TOF-MS were filtered through a 0.22 μm membrane before injection. All the other reagents were of analytical grade and were bought from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China).

Extraction and Characterization of Plant Polyphenols

Methanol and ethanol were from VWR (Darmstadt, Germany) and were like all other solvents used of HPLC grade. Trolox, 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS), Folin-Ciocalteu reagent, quercetin, (−)-epicatechin, procyanidin B1, procyanidin B2, procyanidin C1, p-coumaric acid, trans-cinnamic acid and phloridzin dihydrate were from Sigma–Aldrich (Taufkirchen, Germany). Gallic acid, (+)-catechin, chlorogenic acid, ferulic acid, coffeic acid, rutin trihydrate and acetic acid were purchased from Carl Roth (Karlsruhe, Germany). Avicularin was used from Phytolab (Vestenbergsgreuth, Germany). Protocatechuic acid, hyperoside, isoquercitrin, quercitrin and procyanidin A2 were from Extrasynthese (Genay, France). Reynoutrin was purchased from Carbosynth Limited (Berkshire, UK). 2,2′-Azobis(2-amidinopropane) dihydrochloride (AAPH) was from Fisher Scientific (Nidderau, Germany). HPLC grade water (18 MΩ) was prepared using a MicroPure purification system (Thermo Electron LED GmbH, Niederelbert, Germany). Sodium hydroxide, hydrochloric acid (Merck, Darmstadt, Germany), meta-phosphoric acid (Sigma–Aldrich) and all other chemicals were of analytical grade.

The other standards of caffeoylmalic acid (CAD), procyanidin B2 (PB2), hyperoside (HP), quercitrin (QR), and quercetin 3-O-β-D-(2″-O-β-D-xylopyranosyl)galactopyranoside (QPH) were previously isolated form the leaves of C. bullatus or C. zabelii [50 (link)]. HPLC-grade solvents such as acetonitrile and phosphoric acid were from Avantor Performance Materials (Gliwice, Poland). All other reagents used in the quantitative study were of analytical grade and of the same origin as described elsewhere for the relevant tests [20 (link),21 (link)].

Evaluation of Cocoa Bean Extracts in Rat Diets
