Isolation of Naive B Cells from LN Patients

Peripheral blood mononuclear cells (PBMCs) were isolated from blood samples of patients with LN or healthy controls (HC) by density gradient centrifugation. CD19+ B cells were purified from PBMCs by positive selection using the EasySep™ Human CD19 Positive Selection Kit. Naive B cells were isolated using EasySep™ Human Naive B Cell Isolation Kit (STEMCELL Technologies). Cell purities were checked by FACS (>95%).

Naive B Cell Isolation and Stimulation

PBMCs were purified from blood by density gradient centrifugation. Naive CD27 B cells were isolated with the EasySep Human Naive B cell Isolation Kit (STEMCELL Technologies) following the manufacturer’s instructions. Isolated naive B cells were plated in supplemented IMDM (Thermo Fisher Scientific) as described previously (Warnatz et al., 2009 (link)) at a concentration of 0.15 × 106 cells/ml. Cells were stimulated with trimeric CD40L and IL-21 as described (Warnatz et al., 2009 (link)) in the presence or absence of IL-4 (25 ng/ml; Immunotools).

Isolation of Primary Naive B Cells

Primary naive B cells were obtained from fresh buffy coats provided by the Institute for Transfusion Medicine and Gene Therapy, ITG Freiburg. Peripheral blood mononuclear cells were separated by Ficoll gradient centrifugation of whole blood and negatively selected using EasySep Human Naive B Cell Isolation kit (StemCell). Prior to the experiments, primary naive B cells were controlled for purity and rested overnight. The study was approved by the Institutional Review Board of the University of Freiburg (ethical vote nos. 507/16 and 336/16). Additional details can be found in SI Appendix, Methods.
