Antimicrobial Activity of GA-AgNPs

The antimicrobial activity of the GA-AgNPs against three bacterial strains: S. sanguinis (NCTC 7865), S. mutans (NCTC 10449), L. acidophilus (ATCC 314), and one fungal strain C. albicans (ATCC 10231) was determined using the agar disc diffusion and the microdilution assays. The S. sanguinis and S. mutans were purchased from Davies Diagnostics (Randburg, Johannesburg, South Africa); and L. acidophilus and C. albicans were purchased from the American Type Culture Collection (ATCC; Manassas, Virginia, USA).
All the microbes were cultured on Brain Heart Infusion (BHI) Broth (Sigma–Aldrich), and single colonies were subcultured in BHI agar (Sigma–Aldrich) for all the bacterial strains and Sabouraud dextrose agar for C. albicans at 37°C for 24 hours. Following the overnight incubation, the microbes were adjusted to the 0.5 McFarland standard (Mcf) using DensiCHEK Plus standards (BioMérieux, Inc., Durham, NC, USA).

Evaluation of Probiotic Bacterial Strains

The bacterial species evaluated in this study were B. animalis (from ACTIVIA ® ), B. longum, (ATCC 15707), B. lactis (LMG 18905), and B. dentium (ATCC 27678); L. acidophilus (ATCC 4356), L. casei (ATCC 393); S. mutans (ATCC 25175 and 3VF2), and S. sobrinus (ATCC 27607). All ATCC strains were obtained from the Oswaldo Cruz Foundation (FIOCRUZ, Rio de Janeiro, RJ, Brazil), and the André Tosello Foundation (Campinas, SP, Brazil). S. mutans 3VF2 is a highly acidogenic clinical strain that was previously characterized and also provided by Dr. Renata de Oliveira Mattos-Graner (FOP-UNICAMP, Piracicaba, Brazil). 9 B. animalis was isolated from ACTIVIA ® yogurt. Bifidobacterium, Lactobacillus, and Streptococcus species were isolated from the following media: transgalactosylated oligosaccharides propionate agar supplemented with lithium mupirocin (50 mg/L) (TOS-MUP agar; Merck Millipore, Darmstadt, Germany), Rogosa Agar (Difco Laboratories, Detroit, USA) and Mitis Salivarius Agar (MSA, Difco), respectively. 7 (link) All bacteria were grown at 37 o C for 48 h in a 5% CO 2 atmosphere.

Dairy Microbiome Cultivation and Preservation

We purchased L. bulgaricus (No. 1.1855) and S. thermophilus (No. 1.1854) from China General Microbiological Culture Collection Center. We stored L. plantarum (ATCC 14917), L. acidophilus (ATCC 4356), and L. casei (ATCC 15008) in our laboratory at -80°C in culture plus glycerol (20%, vol/vol) for further use. Raw milk was produced by Ningbo Dairy Group Co., Ltd., and had a fat content of 3.2%. All bacterial strains were cultured at 37°C in de Man, Rogosa, and Sharpe (MRS) medium before milk fermentation.

Antimicrobial Activity of TP1-4 Compounds

A single colony was sub-cultured in Brain Heart Infusion Agar (Sigma Aldrich) for all the bacterial strains and Sabouraud dextrose agar for C. albicans at 37 °C for 24 h. Following the overnight culture, the microbes were adjusted to 0.5 McFarland (McF) standard using DensiCHEK Plus (BioMérieux Inc., Durham, NC, USA).
