Bio plex pro mouse diabetes panel

Manufactured by Bio-Rad
Sourced in United States

The Bio-Plex Pro Mouse Diabetes Panel is a multiplex assay that allows for the simultaneous detection and quantification of multiple analytes related to diabetes in mouse samples. The panel measures a range of biomarkers, providing a comprehensive assessment of the diabetes-related factors in a single experiment.

Quantification of Mouse Serum Metabolites

Serum triglycerides were quantified using a Triglyceride Colorimetric Assay Kit (Cayman Chemicals, Ann Arbor, MI). Metabolically active hormones were quantified using the Bio-Plex Pro Mouse Diabetes Panel (Bio-Rad Laboratories, Hercules, CA). Both were prepared according to the manufacturer’s instructions. Freshly obtained blood samples were allowed to clot for 30 min at room temperature before undergoing centrifugation at 5000 rcf for 5 min to obtain serum samples. We utilized an 8-plex Bio-Plex Pro Mouse Diabetes Panel from Bio-Rad (cat. #171F7001M, Hercules, CA) to quantify ghrelin, GIP, GLP-1, glucagon, insulin, leptin, PAI-1, and resistin in mouse serum.

Plasma Insulin Resistance Markers

Mice were injected I.P. with 10 mg/ml ketamine/xylazine. Blood was collected from the portal vein using an insulin syringe and collected into ETDA-coated microfuge tubes. Blood was centrifuged at 5000 rpm for 10 minutes at 4°C and plasma supernatant was stored at −80°C. Insulin resistance markers and gut peptide hormones were measured using Bio-Rad’s Bio-Plex Pro™ Mouse Diabetes Panel (Hercules, CA) following manufacturer’s instructions.

Serum Protein and Hormone Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Concentrations of several proteins were measured in serum collected at euthanasia. All proteins measured are listed in Table S2. All hormones were measured via a BioPlex Pro Mouse Diabetes Panel (BioRad, Hercules, CA), except insulin-like growth factor (IGF)-1 and vascular endothelial growth factor (VEGF), which were assessed with BioRad singleplex assays. Cytokines were measured via a BioPlex Pro Mouse Cytokine Panel. All assays were read on a BioRad MAGPIX multiplex analyzer.

Serum Metabolic Hormone and Cytokine Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
One week before tumor injection, blood was collected from all mice by submandibular bleed after fasting 4–6 hours, and serum was processed and stored. In 5–8 mice/group, serum concentrations of a panel of metabolic hormones and growth factors was measured using a BioPlex Pro Mouse Diabetes Panel (BioRad, Hercules CA). Adiponectin, insulin-like growth factor (IGF)-1 and transforming growth factor beta 1 (TGF-β1) were measured with BioRad singleplex assays, while a panel of cytokines was measured using a BioPLex Pro Mouse Cytokine Panel (Bio-Rad). A BioRad MAGPIX multiplex analyzer was used to read all assays, as previously described (17 (link)).

Serum insulin and leptin concentrations were measured using the Bio-Plex Pro mouse diabetes panel (BioRad, Hercules, CA, USA). Serum IGF1 concentrations were measured using the IGF1 Mouse Magnetic Luminex Screening Assay (R&D Systems, Minneapolis, MN, USA). Assays were conducted according to manufacturer’s protocols on a Bio-Plex MAGPIX system (BioRad).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
