Nicotine, D2 Receptor Modulation in Neurons

Quinpirole hydrochloride, eticlopride hydrochloride, and gabazine (SR 95531 hydrobromide) were purchased from Tocris Bioscience. (-)-Nicotine hydrogen tartrate salt was purchased from Sigma-Aldrich. 15 min after the first nicotine injection, quinpirole 1 mg/kg was administered intravenously. 5 minutes later, eticlopride 1 mg/kg was administered intravenously. When the neuron activity was returned to baseline, a second nicotine injection was performed. Once D2-R pharmacology was applied, no further neurons were recorded and the animal was discarded.

Electrochemical Measurements in Artificial CSF

All chemicals were purchased from EMD Chemicals Inc. (Darmstadt, Germany) unless otherwise stated. Dopamine hydrochloride, Tris base, Tris hydrochloride, 3,4-ethylenedioxythiophene (EDOT), and GBR-12909 were purchased from Sigma-Aldrich (St. Louis, MO). Eticlopride hydrochloride was purchased from Tocris Bioscience (Avonmouth, Bristol, U.K.). Nafion solution (LQ-1105) was purchased from Ion Power Inc. (New Castle, DE) and used as provided. Ketamine was purchased from Phoenix Pharmaceuticals Inc. (Ketaject, St. Joseph, MO). Artificial cerebral spinal fluid (aCSF) was made by dissolving 15 mM Tris, 126 mM NaCl, 2.5 mM KCl, 20 mM NaHCO3, 2.0 mM NaH2PO4, 1.2 mM Na2SO4, 1.2 mM CaCl2, and 2.0 mM MgCl2 in 18.2 MΩ water (EMD Millipore) and the pH was adjusted to 7.40 using HCl. To produce an aCSF gelatin, porcine gelatin (0.5% by mass) was added and the solution was heated until it dissolved. The mixture was allowed to set at room temperature.

Pharmacological Preparation of Dopamine Agonists

SCH23390 hydrochloride (Tocris Cookson, Bristol, UK) and eticlopride hydrochloride (Tocris Bioscience) were dissolved in 0.9% physiological saline. The drug was intraperitoneally (i.p.) injected at a volume of 1 ml/kg of body weight.

Dopamine Receptor Modulation in Animal Study

SKF 81297 hydrobromide (Tocris Bioscience; D1R agonist) was administered systemically at doses of 0.3 mg/kg and 1.0 mg/kg, and locally at doses of 0.4 µg/µl and 4.0 µg/µl. SCH 23390 hydrochloride (Tocris Bioscience; D1R antagonist) was administered systemically at doses of 0.005mg/kg and 0.01mg/kg, and locally at doses of 0.2µg/µl and 2.0µg/µl. Quinpirole hydrochloride (Tocris Bioscience; D2R agonist) was administered systemically at doses of 0.0125 mg/kg and 0.0375 mg/kg, and eticlopride hydrochloride (Tocris Bioscience; D2R antagonist) was administered systemically at doses of 0.01 mg/kg and 0.03 mg/kg. Doses were calculated as the salt. All drugs were dissolved in 0.9% sterile saline, made in batch, aliquoted, and frozen at -20°C. Individual aliquots were defrosted for use on testing days. 0.9% sterile saline was administered in control sessions. In all experiments, doses were applied in a counterbalanced Latin square approach, although blinding was not used when making up or applying agents.
