Comprehensive Chemical Analysis Protocol

All the chemicals and reagents, including Folin–Ciocalteu phenolic reagent, gallic acid, 2, 2-diphenyl-1 picrylhydrazyl (DPPH), catechin, gallic acid, phenolic acid and flavonoid standards, ascorbic acid, and linoleic acid (75%), were purchased from Sigma-Aldrich Co., (St Louis, MO, USA). All the solvents, butylated hydroxyl toluene (BHT), hydrochloric acid, sodium carbonate, sodium hydroxide, aluminum chloride, and sodium nitrite, were purchased from Merck (Darmstadt, Germany). All chemicals used, including the solvents, were of analytical grade and used without further purification.

Quantifying Total Phenolic Content

Folin-Ciocalteu phenolic reagent (Sigma ® ) was used to determine total phenolic contents of methanolic extracts according to method described by YILDIRIM & al [34] . Calibration curve of gallic acid (Sigma ® ) (0, 50, 100, 150, 200, 250 and 500 mg/L) was prepared as standard. Plant extracts, each concentration of gallic acid and blank were mixed with Folin-Ciocalteu reagent and were neutralized with aqueous sodium carbonate (Na 2 CO 3 ). The mixture was incubated at room temperature for 2 hours. The absorbance of each solution was measured at 765 nm against the blank using the spectrophotometer. Total phenolic compounds in plant extracts were calculated according to the calibration curve of gallic acid. Total phenolic was presented as mg/g gallic acid equivalent (GAE) of dry extract. Three replicates were performed.

Antioxidant Assay Protocol

Folin-Ciocalteu phenolic reagent, anhydrous sodium carbonate, 2,2-diphenyl-1-picrylhydrazyl (DPPH), sodium acetate trihydrate, 2,4,6-tris(2-pyridyl)-s-triazine (TPTZ), ferric chloride hexahydrate, quercetin, potassium persulfate, and 6-hydroxy-2,5,7,8-tetramethylchromane-2-carboxylic acid (Trolox), gallic acid, 2,2-azino-bis(3-ethylbenzothiazoline-6sulfonic acid) (ABTS) were obtained from Sigma-Aldrich products (St. Louis, Missouri, USA). Aluminum chloride and potassium acetate were acquired from Fermont S.A. de C.V. and Meyer S.A. de C.V., Mexico, respectively. All reagent were analytical grade.
