Penicillin-streptomycin, trypsin-EDTA, Fetal bovine serum, RPMI 1640, DMEM-high glucose pyruvate, and Opti-MEM were purchased from Gibco (Grand Island, NY, USA). LPS (O111:B4), ATP and DMSO were purchased from Sigma-Aldrich (St. Louis, MO, USA). Nigericin, nano-SiO2, imiquimod, poly(dA:dT), MSU crystal, Y-VAD-CMK, MCC950, lipofectamine-2000, and mouse IL-1β ELISA kit were purchased from InvitroGen (San Diego, CA, USA). Flagellin was purchased from Adipogen (San Diego, CA, USA). Mouse IL-1β antibody (AF-401-NA) and human IL-1β ELISA kit were purchased from R&D Systems (Minneapolis, MN, USA). Antibodies against mouse caspase-1 (AG-20B-0042-C100), ASC (AG-25B-0006-C100), and NLRP3 (AG-20b-0014-C100) were purchased from Adipogen (San Diego, CA, USA), and mGSDMD (ab209845) was purchased from Abcam (Cambridge, UK). Anti-FLAG antibody (F1804) and β-Actin antibody (SC-4778) were purchased from Sigma-Aldrich (St. Louis, MO, USA) and Santa Cruz biotechnology (Dallas, TX, USA), respectively.

Flagellin and MALP-2 Immunoassay Protocol

High purity flagellin (isolated from Salmonella typhimurium strain 14028) and MALP-2 were purchased from Enzo Life Sciences (USA). For use in ELISA of anti-flagellin antibodies, flagellin was purchased from AdipoGen Life Sciences (Switzerland). LPS from Escherichia coli (026:B6), bafilomycin A1, depleted zymosan, and ovalbumin were purchased from Sigma-Aldrich (USA). CpG-ODN (1826) was from TIB Molbiol (Germany). Biotinylated mouse anti-TLR5 monoclonal antibody (clone ACT5) was a gift from Kensuke Miyake (University of Tokyo, Japan). For flow cytometry, following fluorophore-conjugated antibodies were used: Ly6C-PB (clone HK1.4), Ly6G-APC (clone RB6-8C5) from eBioscience (USA); CD11b-PECy7 (clone M1/70), CD11c-PE (clone HL3) from BD Biosciences (USA); B220-PB (clone RA3-6B2) from TONBO Biosciences (USA); mPDCA1-APC (clone JF05-1C2.4.1) from Miltenyi Biotec (Germany). Following antibodies were used for immunoblotting: IRF3 (clone FL-425) from Santa Cruz Biotechnology (USA); IRF7 (clone EPR4718) from Abcam (UK); phospho-IRF3 (clone 4D4G), phospho-IRF7 (polyclonal) from Cell Signaling Technology (USA). Anti-mouse CD16/32 antibody was purchased from BioLegend (USA). Accudenz was from Accurate Chemical and Scientific Corporation (USA). Horseradish peroxidase (HRP)-conjugated goat anti-mouse Ig antibodies were purchased from Southern Biotech (USA).

Immunoblotting of NLRC4 and Related Proteins

HiPerFect transfection reagent (Qiagen), flagellin (AdipoGen Life Sciences), Lipofectamine 3000 transfection reagent (Thermo Fisher Scientific), TRIzol (Life Technologies), Percoll (Solarbio Life Sciences), gentle cell dissociation and IntestiCult organoid growth medium (STEMCELL Technologies), a pcDNA3.1/V5-His TOPO TA expression kit (Invitrogen), Pierce magnetic RNA-protein pull-down kits (Invitrogen and Thermo Fisher Scientific), and a DIG Northern starter kit (Roche) were purchased. High-throughput screening of natural products against TLR4 signaling

Normocin, Blasticidine, Hygromycin B Gold, QUANTI-Blue, HEK293T-TLR4-MD2-CD14, pNiFty2-SEAP and pNiFty2-56K-SEAP were obtained from InvivoGen, San Diego, CA, USA. LPS were obtained from Sigma-Aldrich, St. Louis, MO, USA. Recombinant human IFN-beta was obtained from Peprotech, Rocky Hill, NJ, USA. Recombinant human CIRP and Flagellin (high purity) were obtained from Adipogen life sciences, San Diego, CA, USA. Mouse S100A13 and PTMA were generated from E coli and then, were subjected to endotoxin removal resin. In both of them the endotoxin concentration was below 1EU per microgram protein. Natural products library was kindly gifted from Dr. Yamada and Prof. Takeda.
