Dopamine HCl and serotonin creatinine sulfate monohydrate were obtained from Sigma-Aldrich. Just prior to use, serotonin was dissolved in artificial sea water (ASW, Instant Ocean Aquarium Systems) and dopamine was dissolved in ASW containing 10 mg% ascorbic acid (ASWA) buffered with sodium bicarbonate (pH 7.0) to retard dopamine oxidation [18 (link)]. The fluorescent probe DiBAC4(3) was obtained from AnaSpec, Inc., and stock and working solutions were stored in brown bottles. DiBAC4(3) stock solutions (24 mM) were dissolved in DMSO and working solutions (2.4 μM) were freshly prepared in ASW. All other chemicals were obtained from Fisher Scientific.

All drugs were administered via intraperitoneal (i.p.) injections except NTI which was injected subcutaneously (s.c.). Cocaine HCl (BDH Ltd., Wellington, NZ) was dissolved in heparinised saline (3.0 U/mL) for intravenous (i.v) infusions and in physiological saline for i.p. injections. The KOPr antagonist nor-BNI (In Vitro Technologies) was dissolved in physiological saline and injected s.c. at a volume of 1 mL/kg 24h prior to experimental testing. Dopamine-HCl (Sigma-Aldrich) was dissolved in Milli-Q water. All drugs weights refer to the salt.

Neurotransmitter Modulation Protocols

All drugs were dissolved in either purified water or dimethyl sulfoxide (DMSO). Stock solutions containing DMSO were made such that the final concentration in slice superfusate was diluted by at least 1000-fold. A769662, dorsomorphin dihydrochloride (compound C), STO609 acetate, and ZD7288 were purchased from Cayman Chemical (USA). Dopamine HCl, GBR12935, cocaine HCl, (−) quinpirole HCl, and (−)-sulpiride were purchased from Sigma–Aldrich (USA). SCH39166 was obtained from Tocris/Bio-techne (UK), and EPPTB (N-(3-ethoxyphenyl)-4-pyrrolidin-1-yl-3-trifluoromethyl-benzamide) was a generous gift from Dr. Aaron Janowsky at VA Portland Health Care System, Portland, OR. A769662 was dissolved to working strength in internal pipette solutions; this agent diffused passively from pipette solutions into the cytosol during whole cell recordings. All other drugs were dissolved to final concentration in aCSF and added to the brain slice superfusate. Typically 60 sec were required for solution changes to traverse the tubing system to show initial effect at the neuron. Dopamine solutions were made daily and kept on ice to retard oxidation.
