Extraction and Identification of Menaquinones

All reagents and solvents used during the extraction were analytical grade. Strains were incubated in 2216E medium (pH 7.4 ~ 7.6), which was composed of 5.0 g/L peptone, 1.0 g/L yeast extract, 0.1 g/L FeC6H5O7, 19.45 g/L NaCl, 5.98 g/L MgCl2, 3.24 g/L Na2SO4, 1.8 g/L CaCl2, 0.55 g/L KCl, 0.16 g/L Na2CO3, 0.08 g/L KBr, 0.034 g/L SrCl2, 0.022 g/L H3BO3, 0.004 g/L Na2SiO3, 0.0024 g/L NaF, 0.0016 g/L NH4NO3 and 0.008 g/L Na2HPO3. Lysozyme (Solarbio Science & Technology, Beijing, China) was used for cell wall digestion. Tris-HCl buffer (10 mM, pH 7.4), chloroform-methanol (2:1 v/v) and hexane-diethyl ether (85:15, v/v) were used for MK extraction. These organic reagents were purchased from Xilong Scientific Co., Ltd., Guangzhou, China. The MK-9 standard was purchased from GLPBIO, Montclair, CA, USA. Mass spectrometry-grade methanol (Sigma-Aldrich, St. Louis, MO, USA) and isopropanol (Fisher Chemical, Thermo-Fisher Scientific Inc.) were used for high-performance liquid chromatography (HPLC), ultra-performance liquid chromatography (UPLC), and mass spectrometer (MS).

Fluconazole Quantification via Mass Spectrometry

Fluconazole (> 99.0%) and its internal standard (IS, Fluconazole-d4 stable isotope (> 99.0%)) were purchased from Toronto Research Chemical (North York, Ontario, Canada). Mass spectrometry-grade methanol, acetonitrile, formic acid, and ammonium formate were obtained from Sigma-Aldrich (St. Louis, MO, USA). Ultra-pure water was obtained using a Milli-Q system (Millipore, Bedford, USA). Stock solutions of Fluconazole (2 mg/mL) and IS (1 mg/mL) were prepared in acetonitrile and stored at − 80 °C in a freezer.

Metabolomics Profiling using UPLC-Orbitrap

Optic Nerve Lipid and Protein Extraction

Single optic nerves were thawed, homogenized via bath sonication and sequential freeze-thaw cycles in dry ice in high salt PBS (1M NaCl) supplemented with a protease-inhibitor cocktail tablet (Roche Diagnostics Corporation, Indianapolis, IN). Phospholipid internal standards dissolved in methanol di-14:0 PE (phosphatidylethanolamine), di-14:0 PC (phosphatidylcholine), 17:0 SM (sphingomylein), 14:0 LPE (lysophosphatidylethanolamine), 14:0 LPC (lusophosphatidylcholine), and di-16:0 PI (phosphatidylinositol) (Materya LLC, Pleasant Gap, PA) were added. The homogenates were then clarified by centrifugation for 15 minutes at 20,000 x g RCF at 4°C. Proteins were precipitated in both the resultant supernatant and pellet by addition of 7 volumes of chilled mass-spectrometry grade methanol (Sigma-Aldrich, St Louis, MO). The methanolic supernatants were combined in a new tube, and the pellets were then extracted with 200 μl of 2:1 chloroform:MeOH to extract lipids. The MeOH and chloroform-MeOH extracts were combined and used for lipid analysis, and the pellets were used for proteomic analyses.

PFAS Adsorption by Chloride Resins

In this study, ten target PFASs (PFBA, PFBS, PFHxA, PFHxS, PFHpA, PFOA, PFOS, PFDA, GenX, and 6:2 FTSA) were purchased from Wellington Laboratories (Guelph, ON, Canada), with their detailed information shown in Tables S1 and S2 in Supplementary Materials (SM). Four types of chloride resins (HPR4700, WA10, S6368, and A111S) were selected. Specifically, HPR4700 (DuPont, Wilmington, NC, USA), WA10 (Mitsubishi Chemical, Tokyo, Japan), S6368 (Lanxess, Cologne, Germany), and A111S (Purolite, King of Prussia, PA, USA) were gel-type strong base resin, gel-type weak base resin, macroreticular strong base resin, and macroreticular weak base resin, respectively. The physical morphology and property of these resins are provided in Figure S1 and Table S3. Fulvic acid (95%) was obtained from Thermo Fisher (Scoresby, North Ryde, Australia). Guaranteed reagents (hydrochloric acid, sodium hydroxide, sodium sulfate, sodium carbonate, and sodium bicarbonate) were all bought from CNW (Shanghai, China). Ultrapure water was produced by the Milli-Q water purification system (Millipore, Boston, MA, USA). Mass spectrometry grade methanol was purchased from Merck (Burlington, MA, USA).
