Modulating MALAT1 and KRAS Expression

MiR-1 mimic and its scramble control (miR-NC), miR-1 inhibitor (anti-miR-1) and its scramble control (anti-miR-NC), and small interference RNA targeting MALAT1 (si-MALAT1) and its scramble control (si-NC) were purchased from Gene Pharma Co., Ltd. (Shanghai, China). The sequences of MALAT1 or KRAS were amplified by polymerase chain reaction (PCR) and subcloned into pcDNA3.1 (Thermo Fisher Scientific) to produce MALAT1 or KRAS overexpression plasmid, named as pcDNA-MALAT1 (MALAT1) or pcDNA-KRAS (KRAS). Then, these miRNAs, miRNA inhibitors, siRNAs, or plasmids were transfected into PC cells using Lipofectamine 2000 (Thermo Fisher Scientific) according to the manufacturer’s instructions.
