Butyl isothiocyanate

Huh-7, Huh7.5, Ava5 (harboring HCV subgenomic replicon; genotype 1b) and Huh7.5/J6/JFHEMCVIRESRlucNeo cells (harboring HCV subgenomic replicon RNA and renilla luciferase reporter gene; genotype 2a) obtained from Apath, LLC (St. Louis, MO) [18 (link)] were routinely passaged in Dulbecco’s modified Eagle’s medium containing 10% heat-inactivated fetal bovine serum, 1% nonessential amino acids, and 1% antibiotic-antimycotic in a 37°C incubator with a humidified atmosphere containing 5% CO₂. Sulforaphane (SFN), phenethyl isothiocyanate, benzyl isothiocyanate, benzyl isothiocyanate, butyl isothiocyanate, allyl isothiocyanate, and HO-1-specific inhibitor (tin protoporphyrin IX dichloride; SnPP) were obtained from Sigma Aldrich Co. (St. Louis, MO, USA). Biliverdin was obtained from MP Biomedicals, Inc (Santa Ana, CA, USA). IFN-α-2a (Roferon-A) was obtained from Roche Ltd (Basel, Switzerland). Daclatasvir and telaprevir were obtained from Legend Stat International Co., Ltd (Omdurman, Sudan). Sofosbuvir was obtained from Shanghai Haoyuan Chemexpress Co., Ltd. (Shanghai, China). These compounds were stored at a concentration of 10 mM in 100% dimethyl sulfoxide (DMSO) at 4°C until use. All the reactions were performed using a final concentration of 0.1% DMSO.

Sulfatase from Helix pomatia, sinigrin hydrate, l-ascorbic acid (AA), trichloroacetic acid (TCA), dl-dithiothreitol (DTT), N-ethylmaleimide (NEM), 2,2′-bipyridyl, iron (III) chloride (FeCl₃), 3-O-CQA (3-O-caffeoylquinic acid), 5-O-CQA (5-O-caffeoylquinic acid), butyl-isothiocyanate, caffeic acid (CA), 1,2-benzenedithiol, ß-mercaptoethanol (HPLC grade), methylene chloride (HPLC grade), sephadex A-25, sephadex C-25, acetonitrile (HPLC grade), methanol (HPLC grade), sulforaphane, sodium acetate, potassium phosphate, and orthophosphoric acid (H₃PO₄) were obtained from Sigma Chemical Co. (St. Louis, MO, USA). Desulfoglucoraphanin was obtained from Santa Cruz Biotechnology (Dallas, TX, USA).

Meloidogyne hapla was maintained on tomato (Solanum lycopersicum) cv. Moneymaker under greenhouse conditions (25/19 °C, 60% humidity, 15/9 h day night cycle). Freshly hatched second-stage juveniles (J2) were extracted from heavily galled root systems placed under a mist chamber (22 °C) [46 ]. Hatched J2 were collected every day and stored at 6 °C until being used in experiments, but no longer than 10 days.
The ITCs allyl isothiocyanate, benzyl isothiocyanate, butyl isothiocyanate, ethyl isothiocyanate, methyl isothiocyanate, phenyl isothiocyanate, and 2-phenylethyl isothiocyanate were obtained from Sigma-Aldrich (St. Louis, MO, USA) and diluted with sterilized water to achieve the concentrations requested in the different experiments. For the allyl isothiocyanate (AITC) experiment in soil, the volumetric measure of the pots was used to calculate the concentration (µmol/mL) applied to the soil.
