Enzymatic Biomass Pretreatment Protocol

by Novozymes A/S, Denmark: an amylase, two serine endoproteases (SE
I and SE II), a cellulase/xylanase, and a peroxidase. Stock solutions
were made in PBS 1× (buffer phosphate solution, pH 7.3–7.5)
from Biopack reagents.
For LB medium preparation, yeast extract
and tryptone were obtained from Britania Laboratories, and sodium
chloride was obtained from Biopack. For M63 minimal medium with 0.4%
arginine, H2PO4, K2HPO4, and MgSO4 were provided by Biopack, (NH4)2SO4 was supplied by Anedra, and l-arginine
free base was provided by Sigma-Aldrich. Deionized water was obtained
by a Double Pass Reverse Osmosis System (Wasserteck, Argentina), with
a resistivity of 1.5 MΩ cm. CV and sugar standards for HPAEC
were provided by Sigma-Aldrich.

Characterization of PTEN DNA Aptamers

PTENz14-106, PTENz7-biotin, PTENz7-106, PTENz7-70, and PS2.M-H1 DNA aptamers were purchased from IDT (Coralville, IA, USA) and Macrogen (Seoul, Korea). Hemin and 3,3’-diaminobenzidine (DAB) were purchased from Sigma-Aldrich (St. Louis, MO, USA). IgG monoclonal PTEN antibodies (sc-20 PTEN Santa Cruz, CA, USA) and HRP-conjugated goat anti-rabbit IgG were purchased from Promega (Madison, WI, USA). Synthetic balsam was purchased from Alwik (Buenos Aires, Argentina). All chemicals were reagent grade and used without further purification. All solutions were prepared with ultra-pure water (Milli-Q).

Pesticides Analysis in Samples

The pesticides standards of deltamethrin, penconazole, kresoxim-methyl, cyproxonazole, epoxiconazole and azoxystrobin were supplied by Sigma-Aldrich (Seelze, Germany). The stock solutions (1000 mg/L) were prepared by dissolving the standards in methanol HPLC grade (99.9%) purchased by Sintorgan (Buenos Aires, Argentina), and stored under freezing condition (-18°C ± 1°C) in dark bottles sealed with PTFE/silicone caps. The working standard solutions (50 mg/L) were prepared in acetonitrile (ACN) of high purity grade for residue analysis, provided by Merck (Darmstadt, Germany).
Deionized water obtained from an E-pure water purification system (Barnstead/Thermolyne, Bedford, MA, United States) was used for sample swelling in the extraction step.
For extraction and partitioning, sodium chloride and anhydrous sodium sulfate were purchased from Biopack (Buenos Aires, Argentina); sodium hydrogencitrate sesquihydrate and sodium citrate dihydrate were obtained from Sigma-Aldrich (Seelze, Germany). For clean-up using d-SPE, C18 and PSA bulk powder were obtained from Agilent Technologies (Santa Clara, United States).

Preparation of Growth Media Reagents

Mueller-Hinton broth, agar, brain-heart infusion, yeast extract, glucose and peptone were obtained from Britania S.A. (Ciudad Autónoma de Buenos Aires, Argentina). Chitosan-Starch Polymer Synthesis

Spray-Drying Encapsulation Protocol

The encapsulating agent used for spray-drying was maltodextrin DE10 (dextrose equivalent 10-MD10) (Ingredion S.A, Argentina). The salts (analytical grade) used to control the relative humidity (RH %) were magnesium chloride (33%), potassium carbonate (43%), sodium bromide (58%), sodium chloride (75%), and ammonium sulfate (80%), purchased from Biopack (Argentina). The Folin-Ciocalteu reagent was obtained from Merck (Darmstadt, Germany). Formic acid (98%), chromatography grade methanol and acetonitrile were purchased from Merck (Darmstadt, Germany). All reactive chemicals were analytical grade or superior. Ultra-pure water was obtained from a RiO/Elix3-Sinergy185 purification system (Millipore, Sao Pablo, Brazil). Cellulose filters (3 μm pore size) and (0.45 μm pore size) nylon membrane were supplied by Microclar (Buenos Aires, Argentina).

Gustatory Heterogeneity Induction Protocol

To generate a gustatory heterogeneity in the arena we added (homogeneously with a micropipette) 100 μl of 1 M NaCl over the filter paper covering the floor of the stimulus zone and 100 μl of distilled water on the control zone (inset Figure 5). The NaCl (purchased in Biopack, Argentina) solution was prepared in distilled water. This concentration was chosen as in previous works it was efficient in deterring feeding in the same species (unpublished data). The experiment started immediately after loading the water and the NaCl solution (i.e., 1 min approximately) in order to minimize water evaporation.
