Quantitative Lung Tissue Immunohistochemistry

Check if the same lab product or an alternative is used in the 5 most similar protocols
Lung tissue was analyzed for detection of PCNA (2586, Cell Signaling) and MMP-9 (13667, Cell Signaling) as published (Perer et al., 2020 (link); Jandova and Wondrak, 2021 (link)). Nuclear staining was performed using Hematoxylin QS counterstain (H3404; Vector Laboratories, Burlingame, CA). Images were captured using an Olympus BX50 and Spot (Model 2.3.0) camera. Quantitative tissue scoring (H-score) was performed as published (Bair et al., 2010 (link)).

Cell Migration and Invasion Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
In all, 3 × 104 or 5 × 104 cells were suspended in serum-free media in 8-μm transwell inserts (#83.3932.800, Sarstedt), for 24-h migration or 48-h invasion assays, respectively. For the invasion assay, the upper chamber was pre-coated with 1/9 dilution of Matrigel (Cultrex Reduced Growth Factor Basement Membrane Extract, #3433-005-01, R&D Systems). After incubation, inserts were washed with PBS. Migrated or invaded cells on the underside of the membrane were fixed with 4% paraformaldehyde and stained with hematoxylin (Hematoxylin QS Counterstain, #H-3404, Vector Laboratories). Migrated or invaded cells were counted from six representative fields from the inverted EVOS XL core microscope (Thermo Fisher Scientific).

Frozen sections were treated with Hematoxylin QS Counterstain (#H-3404-100, Vector Laboratories), followed by three washes in distilled water. Sections were then treated briefly with bluing buffer (#CD702, DAKO), followed by three washes in distilled water. Finally, sections were treated briefly with Eosin Y solution (#HT110116, Sigma) followed by three washes in distilled water.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
