Cultivation Media Preparation for Microalgae

Cultivation of Bacteroides thetaiotaomicron

B. thetaiotaomicron was routinely grown on brain- heart infusion (BHI, OXOID) supplemented with hemin (1 μg/ml, Sigma-Aldrich). Agar (Fisher bioreagents), erythromycin (25 μg/ml, Duchefa Biochemies), gentamicin (200 μg/ml, Formedium) and 5-fluoro-2′-deoxyuridine (FUdR, 200 μg/ml, Sigma-Aldrich) were added when required. Minimal medium (MM) contained 7.5 mM NH4SO4 (Thermo Scientific), 9.5 mM Na2CO3 (Melford), 4 mM L-Cysteine (Melford), 100 mM KH2PO4 (pH 7.2)(Fisher Chemical), 1 μg/ml menadione (Sigma-Aldrich), 1 μg/ml hemin and a mixture of mineral salts (15 mM NaCl (Duchefa Biochemies), 0.18 mM CaCl2•2H20 (Fisher Chemical), 0.1 mM MgCl2•6H2O (Fisher Chemical), 0.5 mM MnCl2•4H2O (Acros Organics) and 0.04 mM CoCl2•6H2O (Sigma-Aldrich)). Vitamin B12 (Sigma-Aldrich) and L-methionine (Formedium) were added as required and described in the text. 0.5% of fructose (Thermo scientific) was added as carbon source. Cells were grown in a Whitley A35 anaerobic workstation with a mixture of gas of 80% N2, 10% CO2 and 10% H2.

Bovine Skin Gelatin-based Cell Culture

CuSO4×5H2O, MnCl2×4H2O, CoCl2×6H2O, glutaric dialdehyde (GDA), p-formaldehyde, β-nicotinamide adenine dinucleotide (disodium salt hydrate), sodium pyruvate, sodium borohydride were produced by Acros Organics. Citrus pectin (classic CM 201) was produced by Herbstreith&Fox.
Anti-Cytokeratin 10 antibody [EP1607IHCY] (Alexa Fluor® 647) (Abcam, ab194231) and CellTracker Green CMFDA Dye (Thermo Fisher Scientific), isoflurane (Abbott Laboratories) were used. Histological stains were obtained from BioVitrum (Russia). Reagents and consumables for cell culturing were purchased from Paneco (Russia). Ultrapure water (18.2 MΩ×cm, Milli-QAdvantageA10, MerckMillipore) was used to prepare aqueous solutions and buffers.

Synthesis of Mn- and Yb-Doped Zeolite-Y

Powder samples of Mn2+ doped zeolite-Y (denoted as xMn/ zeolite-Y, x = 0.4, 0.8, 1.2 mol/L), Yb3+ doped zeolite-Y (denoted as yYb/zeolite-Y, y = 0.05, 0.1, 0.2, 0.4, 0.8 mol/L), and those codoped with both Mn2+ and Yb3+ (denoted as yYb3+, xMn2+/zeolite-Y, y = 0.2 mol/L, x = 0, 0.2, 0.4, 0.6, 0.8, 1.0 mol/L) were prepared by ion-exchange reaction method. The raw materials zeolite-Y (SiO2/Al2O3 = 5.1), Yb(NO3)3·5H2O (99.9%) and MnCl2·4H2O (99.99%) were purchased from Alfa Aesar. The zeolites were initially stirred in the aqueous solution of MnCl2·4H2O and Yb(NO3)3·5H2O at 80 °C for 36 h. Then the resulting products were centrifugated and washed with deionized water. After several times of washing and centrifugation, the products were kept in the oven at 80 °C for 12 h for drying. Finally, they were placed into an alumina crucible and sintered at 800 °C for 1 h under a reductive atmosphere (5%H2 + 95%N2) in a furnace.

Optimized Bacterial Culture and IBA Production

Overnight cultures were grown in 5 ml Luria broth media containing appropriate antibiotics. Antibiotic concentrations were as follows: kanamycin (50 μg ml−1; IBI Scientific), ampicillin (200 μg ml−1; Fisher BioReagents), tetracycline (20 μg ml−1; Fisher BioReagents) and spectomycin (50 μg ml−1). IBA production was carried out with M9P medium, consisting of M9 medium (33.7 mM Na2HPO4, 22 mM KH2PO4, 8.55 mM NaCl, 9.35 mM NH4Cl, 1 mM MgSO4 and 0.1 mM CaCl2; BD Bacto); 5 g l−1 yeast extract (BD Bacto); 50 g l−1 glucose (Fisher BioReagents); and 1,000-fold dilution of A5 trace metal mix (2.86 g H3BO3 (Fisher Chemical), 1.81 g MnCl2·4H2O (Alfa Aesar), 0.079 g CuSO4·5H2O (Sigma-Aldrich) and 49.4 mg Co(NO3)2·6H2O (Sigma-Aldrich) per liter water). Optical densities (OD) were measured at 600 nm with a Synergy H1 hybrid plate reader (BioTek Instruments, Inc.).
