Dermal Fibroblast Culture and Stimulation

DHLFs were purchased from Lonza (Basel, Switzerland) and cultured in fibroblast growth medium (FBM, Lonza, Walkersville, MD, USA). Recombinant human TGFβ and PDGF were obtained from Peprotech (Rocky Hill, CT, USA) and used at a final concentration of 5 ng/ml. Chemically synthesized ONG41008 was obtained from Syngene International Ltd. (Bangalore, India), dissolved at a stock concentration of 50 mM in DMSO, and stored in aliquots at −20 °C. The remaining 32 CDs in the test set were purchased from Analyticon (Berlin, Germany). For simultaneous treatment, ONGHEPA1 cells (5.5 × 104 cells/well in 1 ml) were seeded in 24-well plates. The anti-fibrotic effect of eupatilin was examined under a microscope (EVOS, Life Technologies, Carlsbad, CA, USA).
