Immunoblotting of NLRP3 and Caspase-1

A 10 μg of protein from lung tissue sample was electrophoresed on 15% polyacrylamide gels using a discontinuous system. The proteins were transferred to nitrocellulose membranes at 120 V for 40 minutes. After blocking in 5% skim milk and 0.1% NP40 in TBS for 2 hours at room temperature, the membranes were incubated with a rabbit polyclonal anti-mouse NLRP3-inflammasome (Abnova) or rabbit monoclonal anti-mouse caspase-1 (Abcam, Cambridge, MA, USA) primary antibody. The membrane was incubated with horseradish peroxidase (HRP)-conjugated anti-rabbit immunoglobulin G (1:2,000 dilution) for 1 hour at room temperature. The target protein was detected using an enhanced chemiluminescence reagent (Amersham Pharmacia Biotech, Little Chalfont, UK).
