Tnf α elisa kit

Manufactured by R&D Systems
Sourced in United States, United Kingdom, Japan, China

The TNF-α ELISA kit is a laboratory tool used to detect and quantify the cytokine tumor necrosis factor alpha (TNF-α) in biological samples. This kit employs the enzyme-linked immunosorbent assay (ELISA) technique to measure the concentration of TNF-α present in the sample.

Bronchial Epithelial Cell Culture and Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Bronchial epithelial cell growth medium (BEGM) with antibiotics was purchased from Lonza (Walkersville, MD). The BEGM was prepared following manufacturer’s guideline, which contained all the supplements (BPE, hEGF, epinephrine, transferrin, insulin, retinoic acid, triiodothyronine, GA) except hydrocortisone to avoid any inhibitory effect of corticosteroids on cell pro-inflammatory responses. RNA lysis buffer RLT was from Qiagen (Hilden, Germany). RIPA western lysis buffer was purchased from Thermo-Fisher Scientific (Waltham, MA). DMEM (high glucose) for making D10 (DMEM + 10% FBS + 1% Pen/Strep + 1% Amphotericin B + 1% L-Glutamine+ 0.5% Gentamicin) was from GE Life Sciences (Logan, UT). The nuclear extraction kit and TransAM NF-κB p65 assay kit were from Active Motif (Carlsbad, CA). IL-8, IP-10 and TNF-α ELISA kits were obtained from R&D systems (Minnieaplois, MN).

Cytokine Quantification in RASF

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cytokine concentrations from the harvested supernatants of RASFs after appropriate treatment were measured by using commercial interleukins IL-6, IL-1β and IL-8 and tumor necrosis factor-α (TNF-α) ELISA kits (R&D Systems, Minneapolis, USA) according to the instructions of the manufacturer. Each sample was prepared in triplicate, and the experiment was repeated three times.

Blood was collected retro-orbitally under anesthesia immediately prior to sacrifice. Serum was obtained using serum separator tubes with lithium heparin (Becton Dickinson) and kept at -80C. ELISA kits were purchased from: Mouse adiponectin, leptin, adipsin, resistin and TNFα ELISA kits from R&D; Mouse Viafatin and Insulin ELISA kit from Ray biotech (Norcross, GA); Serum CTx-1 and TRAP5b kits from Nordic Bioscience; osteocalcin ELISA kit from Biomedical Technologies Inc.

Cytokine Expression in Vascular Tissues

Check if the same lab product or an alternative is used in the 5 most similar protocols
Interleukin‐1β, IL‐6, and TNF‐α ELISA kits (purchased from R&D System) were performed to detect the expression of inflammatory cytokines of vascular tissues of circles of Willis. Tissues were treated according to the manufacturer's protocols, and the supernatant was added into 96‐well plates coated with primary antibodies. After washing four times, biotinylated antibodies were added into each well for 30 min at 37°C, and detected using horseradish peroxidase (HRP)‐conjugated streptavidin and chromogen reagent. Absorbance at 570 nm was immediately detected using an ELISA reader (Molecular Devices).

Cell culture supernatants from in vitro experiments were harvested and used to measure IL-2, IFNγ, and TNFα by ELISA. Antibody pairs for IFNγ and IL-2 ELISAs were purchased from BD Biosciences, and TNFα ELISA kits were purchased from R&D according to the manufacturer’s instructions. ELISAs were read on a SpectraMax M2 microplate reader (Molecular Devices) and data analyzed using SoftMax Pro version 7.0.2 software (Molecular Devices). Quantifying Cytokine Production in BMDMs

Check if the same lab product or an alternative is used in the 5 most similar protocols
To measure the production of cytokine, BMDMs were stimulated with Clostridium perfringens (MOI = 5). An equal amount of PBS was added to the uninfected group. After 1 h, cells were treated with RPMI 1640 medium containing 30 μg/mL gentamicin, 200 U/mL penicillin, and 200 μg/mL streptomycin to kill extracellular bacteria. After 1 h, discard the culture medium and replace it with RPMI 1640 culture medium containing 10 μg/mL gentamicin and 100 U/mL penicillin, and 100 μg/mL streptomycin to continue culturing for 22 h. The cell supernatants were collected and used for inflammatory cytokine examination with TNF-α ELISA kits following the R&D Systems (DY410) instruction.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
