Husbandry of Transgenic C57BL/6 Mice

Animal care and use were conducted according to established guidelines approved by the Institutional Animal Care and Use Committee at Wenzhou Medical University (Approval number: 2018-211). Wild type C57BL/6 mice, purchased from Nanjing Medical University, were housed at 20–25°C in a light-controlled (12/12 h) room with unrestricted access to water and food. Transgenic green fluorescent protein (GFP) C57BL/6 mice were originally obtained from Jackson Laboratory and raised in our facility.

Murine Microsurgical Ischemic Model

C57BL/6 and GFP+ C57BL/6 mice were obtained from Jackson Laboratories. Male mice, 12-week-old weighing 25–30g were used. Surgical procedures were carried out under an operative microscope (Carl Zeiss, Thornwood, NY), with 10× to 40× magnifications (Supplemental video). The microsurgical instruments used in this model are listed in Supplemental Table 1. All animal procedures in this study were conducted in accordance with experimental protocols that were approved by the Institutional Animal Care and Use Committee at the University of Wisconsin, Madison (Protocol M02284).

Murine Model for Genetic Analyses

Eight- to 12-week-old, sex- and age-matched wild type (WT, C57BL/6) or DiYF mice were used in this study. Seven generations of back crossed GFP+/+-C57BL/6 mice were purchased from The Jackson Laboratory (Bar Harbor, ME). GFP+/+-DiYF mice were bred from DiYF and GFP heterozygous mice. All animal procedures were performed in accordance with an approved institutional protocol according to the guidelines of the Institutional Animal Care and Use Committee of the Cleveland Clinic.
