saline (PBS) tablets, silver nitrate (≥99.0%), gold(III) chloride
trihydrate (≥99.9%), sodium citrate dihydrate (≥99.0%), l-ascorbic acid (≥99.0%), potassium chloride (≥99.0%),
sulfuric acid (98%), methanol (≥99.9%), polyethylenimine (PEI,
MW ≈ 25000 by LS), Nafion solution (5 wt %), and 4-aminothiophenol

bacterial glucose dehydrogenase (GDH) from Ewingella

was purified and characterized according
to a previously published protocol.39 (link),45 (link) All aqueous
solutions were prepared in ultrapure water (resistivity of 18.2 MΩ
cm) purified by a Purelab Flex system (ELGA LabWater, High Wycombe,
UK). For whole blood analysis, blood samples from healthy donors were
collected in VACUETTE FC Mix tubes (Greiner Bio-One, Kremsmünster,
Austria). The tubes contain a mixture of additives Na2EDTA,
sodium fluoride, citric acid, and sodium citrate, which inhibits glycolysis
and prevents coagulation. Blood collection for analysis with the biosensors
was approved by the Swedish Ethical Board (approval number 2009/180).

D-Lactate Analysis from Blood Samples

Blood samples for D-lactate analysis were collected in 3-mL Vacuette® FC-mix tubes (Cat # 454513; Greiner Bio-One International GmbH), together with routine blood samples according to the protocol for blood sampling in the routine setting. The samples were drawn at admission after written and verbal consent were obtained according to the approval from the North Denmark Region Committee on Health Research Ethics. High-risk patients were included when AIN was suspected, and blood samples were collected in 3-mL Vacuette® FC-mix tubes after written and verbal consent were acquired and prior to surgery, in accordance with the above-mentioned approval.

Standardized Glucose Measurement Protocol

The third sample was collected into a VACUETTE® FC Mix Tube (Greiner Bio-One, containing a lyophilized mixture of Na2EDTA, sodium fluoride, citric acid, and sodium citrate) and was kept at room temperature until analysis within 3–4 h. The midwife delivered the samples to the laboratory directly after completion of the OGTT. Glucose was analyzed on a Cobas 8000 modular analyzer (Roche Diagnostics Ltd., Rotkreuz, Switzerland) by a hexokinase method (hereon referred to as CL) traceable to the glucose ID GC-MS. The method has shown a mean imprecision (CV%) of 1.0 and 1.1 at plasma glucose levels 3 mmol/L and 20 mmol/L, respectively (n = 50). The laboratory is accredited according to SS-EN ISO 15189 and participates in interlaboratory comparison schemes from Equalis (https://www.equalis.se/en/).
All women had their BMI calculated at their first antenatal visit. Clinical and sociodemographic details were recorded at the time of the diagnostic OGTT at the hospital.

Fasting Blood Sample Collection

Blood samples were collected between 7.45 and 8.30 in the morning after 10 hours of fasting. Venous blood samples were collected with minimal stasis after 15 minutes rest in a supine position. The first 5 mL collected were discarded. The following 4 mL were collected in clot activator tubes (Becton-Dickinson, Plymouth, UK; BD Ref: 369032) and used for analyses of insulin and CRP in serum. Next, 3 mL were collected in trisodium citrate tubes (0.109 mol/L Na 3 Citrate, BD Ref: 363048) for analysis of vWF and t-PA:Ag. Finally, 3 mL were collected in EDTA-tubes (K 2 -EDTA: 5.4 mg, BD Ref: 367525) for IL-6 and ICAM-1, and 3 mL were collected in EDTA-tubes with citric acid and sodium fluoride (Vacuette FC Mix Tube, Greiner Bio-One, Frickenhausen, Germany, Ref: 454513) for plasma glucose measurements. Immediately after sampling, platelet poor plasma was prepared by centrifugation for 20 minutes at 2000 x g (20°C). Plasma and serum were transferred to aliquots, rapidly frozen and stored at -80 °C until testing.
