Immunohistochemical Analysis of Aortic Sinus

The paraffin-embedded aorta sinus sections (4 μm) were deparaffinized for citrate or EDTA antigen retrieval according to the manufacturer's instructions, blocked with hydrogen peroxide (H2O2, 3%), and incubated with different primary antibodies: CD68 (Abcam, ab125212, 1 : 500), TNF-α (Abcam, ab66579, 1 : 200), IL-6 (Abcam, ab9324, 1 : 300), NF-κB (Santa Cruz, sc-8414, 1 : 500), Nrf2 (Abcam, ab137550, 1 : 300), and HO-1 (Abcam, ab13243, 1 : 500) for 60-minute incubation at 37°C. Finally, the sections were washed again with PBS for DAB colorimetry. The positive sections were quantified by Image-Pro Plus 6.0 software (NIH, Bethesda, MD, USA).
