Urine samples were collected overnight (approximately 16 h) using a cage pan, once during the acclimation period, on days 31 and 59. Animals were not fasted prior to blood collection. The collected urine samples were centrifuged (1500 rpm, 5 min at room temperature) for urine chemistry and urine sediment analysis. Urinalysis parameters were measured by reading the scales on the urine collection tubes of Combur 10 TM urine sticks (Roche) and Cobas U411 urine analyzer (Roche) and then examined visually or under a microscope. Urine potassium (UK), chloride (UCl), and sodium (Una) levels were measured using a Toshiba 2000FR NEO chemistry analyzer.

Overnight Urine Analysis in Fasted Animals

Urine was collected overnight (∼ 16 h period) the day before terminal sacrifice. Animals were housed in individual metabolic cages during this period. Water was available ad libitum but animals were fasted. Urine volume was measured and the following parameters were analyzed using a Cobas U411 uring analyzer (Roche, Germany) and Combur 10 T M urine sticks (Roche, Germany): pH, specific gravity, bilirubin, protein, urobilinogen, nitrite, glucose, erythrocytes and ketones. Urine cast, epithelial cells, white blood cells and red blood cells were examined by microscopy of urine micro sediments.

Comprehensive Urinalysis Protocol
