Immunofluorescence Staining of DNA Damage Proteins

Cells grown on coverslips were prepared for IF by standard procedures. Telomere FISH was performed subsequent to antibody incubation using Telomere PNA FISH Kit/Cy3 (Dako K5326).

Western Blot Analysis of Chromatin Regulators

Cells were lysed with RIPA buffer supplemented with protease inhibitor cocktail and PMSF. A quantity of 50 μg cell lysate was loaded into a SDS–PAGE gel and transferred onto a polyvinylidene difluoride membrane (Bio-Rad). The membrane was blocked with 5% skim milk at room temperature for 1 h, followed by overnight incubation with primary antibodies at 4 °C. The blot was subsequently incubated with either horseradish peroxidase (HRP)-conjugated anti-mouse IgG, HRP-conjugated anti-rabbit IgG, or HRP-conjugated anti-goat IgG (1:10,000, Beith). Signal was detected using SuperSignal West Dura Extended Duration Substrate (Thermo Scientific) and captured on CLXposure film (Thermo Scientific).
