electrode puller (Zeitz-Instruments Vertriebs GmbH, Munich, Germany)
followed by gold coating with a SC7640 sputter coater (Quorum Technologies,
Kent, UK).

Optimized Protein Denaturation and Digestion

Denatured samples were applied to pre-wet Vivacon Hydrosart® stabilized cellulose filters (30KDa molecular weight cutoff; Sartorius) connected to LoBind tubes for reduction and alkylation steps. Samples were reduced with 20 mM dithiothreitol (DTT; Sigma) in 8 M Urea buffer for 30 min at 37 °C, followed by alkylation of free cysteines with 50 mM iodoacetamide (IAA; Sigma) in 8 M urea buffer for 20 min at room temperature (RT) in the dark. The flowthrough was discarded, and filters were washed three times with 2 M urea in 50 mM ammonium acetate, pH 5.5 (2 M urea buffer) before digestion for 3 h at 37°C using rLys-C and trypsin from the AccuMAPTM Low pH Protein Digestion Kit (Promega). Tubes were subsequently centrifuged to collect tryptic peptides in the flowthrough. 20% acetonitrile (Biosolve) in 50 mM ammonium acetate pH 5.5 was added to the filter to reduce non-specific binding followed by a further centrifugation step. Finally, 1% acetic acid in LC-MS grade water (Biosolve) was added to the flowthrough to inactivate enzymes prior to storage of the sample at −80 °C until solid-phase extraction and analysis by LC-MS/MS.

Decitabine Formulation Development

Decitabine was purchased from LC Laboratories® (Woburn, MA, USA). Labrafac® WL1349 (caprylic-capric acid triglycerides), Labrafil® M1944CS (oleoyl macrogol-6 glycerides), Peceol® (glycerol mono-oleates), and Transcutol® HP (THP) (highly purified diethylene glycol monoethyl ether) were kindly provided by Gattefossé (Saint-Priest, France). Sodium phosphate dibasic dodecahydrate, potassium dihydrogen phosphate, formic acid, ammonium acetate, Triton® X-100, Tween® 80, Span® 80, and formic acid were obtained from Sigma-Aldrich (St Quentin-Fallavier, France). Captex® 8000 (glyceryl tricaprylate) was a gift from Abitec Corp. (Saint-Quentin Fallavier, France).
Absolute ethanol, methanol ULC/MS grade, acetonitrile, and LC-MS grade water were purchased from Biosolve (Dieuze, France). Acetone was purchased from VWR (Fontenay-sous-Bois, France).

Peptide Synthesis with HBTU and Fmoc-Protected Amino Acids

Trifluoroacetic acid (TFA) was obtained from Biosolve. Decane (synthesis grade) was purchased from Merck. Solvents and reagents were used as received. Water was purified with a Milli-Q UltraPure Water Purification System.

Enzymatic Saccharification of Rice Straw

All the chemicals used in this study were procured from Sigma Aldrich and Merck, India.
13C gluconic acid (99 atom % 13C) were purchased from Omicron Biochemicals, USA, and LC/MS grade water was obtained from Biosolve Chimie, SARL, France. Commercial enzymes Cellulase from T. reesei ATCC 26921 (Celluclast 1.5L) and β-glucosidase (Novozymes 188) were procured from Sigma, Aldrich India. Cellic® Ctec2 and Cellic® HTec3 were a kind gift from Novozymes, Mumbai, India. Avicel® PH 101 and dilute acid-treated rice (Oryza sativa) straw were used as substrates for enzymatic saccharification studies. Rice (Oryza sativa) straw was procured from the local market at Mathura (27.28° N, 77.41° E) in Uttar Pradesh (North, India) air-dried, ground to the particle size of ~ 10 mm using a knife mill ,and stored in an air tight container for further use. All the experiments were conducted from the single stock of harvested rice straw.

Solvents and reagents were used as received.
