PHGDH Enzyme Activity Assay

The enzyme assay was performed as previously reported²¹ (link). Briefly, PHGDH activity was measured in 96-well plates at 37 °C by monitoring resorufin fluorescence (Ex 550 nm/Em 580 nm) over time with a FLUOstar Omega (BMG Labtech). PSAT1 was included to prevent product inhibition of PHGDH. Assays were performed in the PHGDH assay buffer (50 mmol/L Tris, pH 8.5, and 1 mmol/L EDTA). Substrates and enzyme concentrations were as follows: 3-PG (Sigma–Aldrich), 240 μmol/L; NAD⁺ (Sigma–Aldrich), 120 μmol/L; glutamate (Yuanye Bio-Technology), 30 mmol/L; resazurin (Yuanye Bio-Technology), 0.1 mmol/L; PHGDH (Zoonbio Biotechnology), 15 ng/μL; PSAT1 (Zoonbio Biotechnology), 20 ng/μL; diaphorase (Yuanye Bio-Technology), 0.18 μg/μL. For inhibition assay, compounds and enzyme were preincubated for 30 min before initiating the enzyme reaction with substrate mix.

(Shanghai, China). Acetonitrile (chromatographically pure) was purchased from Macklin Biochemical Co. Ltd. (Shanghai, China). All other chemicals were analytical reagent grade.
