Quantifying Luciferase Activity via D-luciferin

Metastasis Tracking in Nude Mice

Animal experiments were approved by the Animal Care and Use Committee of Zhejiang University. Male nude mice, 4–6 weeks old, were purchased from Shanghai SLAC Laboratory Animal (Shanghai, China). Luciferase-expressing HCT116 cells transduced with lentivirus-expressing sh-control or sh-AAMP (1 × 106 cells/mouse) were injected via the tail vein. D-Luciferin (15 mg/mL, 150 mg/kg) obtained from Meilunbio (Dalian, China) was injected intraperitoneally into mice 15 min before imaging. Metastasis was monitored by bioluminescence imaging using the IVIS imaging system (PerkinElmer, USA). Endpoint assays were conducted 8 weeks after inoculation.
