Western Blot Analysis of Protein Expression

Total protein was extracted from lung tissues or MRC-5 cells using RIPA lysis buffer (KeyGEN, China) containing proteinase and phosphatase inhibitor. The protein concentration was measured with the BCA protein assay kit (Solarbio, USA). Aliquots of 20-40 µg proteins were separated with 8%, 12%, or 15% SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and transferred to PVDF membranes (Millipore, USA). After blocking with 5% skim milk (Biofroxx, Germany) or bovine albumin (Solarbio, USA), membranes were incubated with specific primary antibodies overnight at 4℃, then incubated with secondary antibodies at RT for 1.5 hours. Finally, an enhanced chemiluminescence detection kit (KeyGEN, China) was used to detect proteins and the ImageJ software was used for the grayscale analysis of protein bands. The antibodies used for western blotting are also listed in Table S1.

Ltd. Additionally, human serum was obtained from the First Affiliated Hospital of Zhengzhou University after informed consent of the patient. This research protocol was in accordance with the ethical standards of the 1964 Declaration of Helsinki and its subsequent revisions and was approved by the ethics committee of the First Affiliated Hospital of Zhengzhou University.

Pumpkin Peel Chromium Extraction and Evaluation

(Shanghai, China). Sodium acetate, anhydrous (electrochemical grade), and SS254-500 sodium hydroxide solution were purchased from Thermo Fisher Scientific (Sunnyrale, CA, USA). High-glucose DMEM medium was obtained from Wuhan Servicebio Technology Co., Ltd. (Wuhan, China). Fetal bovine serum was purchased from Zhejiang Tianhang Biotechnology Co., Ltd. (Huzhou, China). Trivalent chromium standard solution was supplied by Beijing Century Aoke Biotechnology Co. Ltd. (Beijing, China). All other chemicals and reagents used were of analytical grade.
